Expression of Hr4Scer\UAS.cQa under the control of Scer\GAL4P0206 results in first instar larval arrest. These L1 larvae grow very large, accumulate lipids in their fat bodies, and have larger organs than controls due to continued proliferation. These flies also exhibit small ring glands compared to controls. This indicates that expression of Hr4 in the prothoracic glands specifically blocks molting and does not trigger lethality.
Expression of Hr4Scer\UAS.cQa under the control of Scer\GAL4phm.PO results in second instar larval arrest. Prepupae form with an approximate 20 hour delay, compared with controls.
Addition of 20E to the feeding medium rescues the lethality found upon expression of Hr4Scer\UAS.cQa under the control of Scer\GAL4P0206 or Scer\GAL4phm.PO.
Hr4UAS.cQa, Scer\GAL4P0206 has abnormal developmental rate | second instar larval stage phenotype, suppressible by Ras85DV12.UAS, Scer\GAL4P0206
Scer\GAL4P0206/Hr4UAS.cQa is a suppressor of abnormal developmental rate | larval stage phenotype of Ras85DV12.UAS, Scer\GAL4phtm.PO
Scer\GAL4P0206/Hr4UAS.cQa is a suppressor of decreased body size | pupal stage phenotype of Ras85DV12.UAS, Scer\GAL4phtm.PO
Hr4UAS.cQa, Scer\GAL4P0206 has ring gland phenotype, suppressible by Ras85DV12.UAS, Scer\GAL4P0206
Scer\GAL4P0206/Hr4UAS.cQa is a suppressor of prothoracic gland phenotype of Ras85DV12.UAS, Scer\GAL4phtm.PO
Co-expression of Ras85DV12.Scer\UAS and Hr4Scer\UAS.cQa under the control of Scer\GAL4P0206 results in wild-type timing of pupariation, and a partial suppression of the Hr4Scer\UAS.cQa-mediated lethality. Hr4Scer\UAS.cQa co-expression with Ras85DV12.Scer\UAS represses the hyper-proliferation of the ring gland seen in Ras85D overexpression flies.
