Expression of Scop\tra2Scer\UAS.cMa in the tarsal region of the foreleg imaginal disc, under the control of Scer\GAL4rn-GAL4-5 in XX pseduomales and in their brother XY males, both mutant for tra2B, Df(2R)trix and Scer\GAL80ts.αTub84B results in a number of defects. The foreleg basitarsus contains several transversal rows, the last one forming the sex comb structure in males and in xx pseudomales mutant for tra2B. This is composed of dark, thick bristles, and is rotated to lie prallele to the proximal-distal leg axis. females lack the sex comb. A significant reduction is seen in the number of bristles forming the male sex comb structure in the foreleg basitarsus of XX pseudomales raised at 25[o]C compared to those raised at 18[o]C. The sex comb size of these latter pseudomales is the same as the sex comb of their XY brothers whether raised at 18 or 25[o]C. Thus, the Scop\tra2 protein supplies tra2 function in Drosophila.

Df(2R)trix, tra2B/tra2B XX pseudomales carrying Scop\tra2Scer\UAS.cMa under the control of the heat-shock inducible Scer\GAL4hs.PB exhibit signs of feminization.
