Expression of Adf1S64A.S184A.Scer\UAS.T:Zzzz\FLAG in the RP2 motor neurons (using the Scer\GAL4eve.RRK driver to drive expression of Scer\FLP1Scer\UAS.cUa which then induces clones of cells expressing Scer\GAL4Act.PU) results in a dramatic decrease in dendritic complexity, number of dendritic branches and total dendrite length compared to controls.
The amplitude of the excitatory junctional potential (WWJP) and the miniature EJP are normal in animals expressing Adf1S64A.S184A.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4futsch-C380. mEJP frequency and quantal content are also unaffected. 50% of the animals fail to display motor neuron firing following stimulation of contralateral sensory afferents at 10Hz for 1.5 seconds. This defect is still seen if Scer\GAL80Cha.PK is also present.
Larvae expressing Adf1S64A.S184A.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4futsch-C380 show reduced locomotion compared to controls.
Weak expression of Adf1S64A.S184A.Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4futsch-C380 results in adults which show prominent defects in the Buridan assay. These defects are still seen if expression is limited to the adult stage using Scer\GAL80ts.αTub84B.
