UAS regulatory sequences drive expression of a full-length Lmx1a open reading frame (ORF) that is tagged at the C-terminal end with three copies of the Tag:HA epitope. The ORF is flanked by a pair of incompatible FRT sites (FRT5 and FRT2), which allows for future in vivo exchange of either the promoter or tag sequence.

Expressing Lmx1aUAS.ORF.GW.Tag:HA under teh control of Scer\GAL4GMR42B09 significantly restores overall ovary morphogenesis, including sheath development, TF formation and egg laying in Lmx1aΔ1 homozygous females. Ovary morphology is also partially rescued if this expression is only induced from mid-L3 but not from 2h after pupariation (using Gal80[ts] to temporally control the expression).
