Approximate extent of Clk rescue fragment inferred from primer sequences.
Clkout homozygotes bearing ClkTag:V5.WT that are kept under constant darkness and subject to 12h:12h 24[o] C:29 [o]C temperature cycles exhibit bimodal peaks of activity with an anticipatory activity increase before temperature transitions; upon transfer to constant 24[o]C, these individuals maintain some rhythmicity and manifest a 23.1h period.
A slightly shortened circadian period and little effect on the overall rhythm strength is observed in flies with one extra copy of ClkT:SV5\V5.WT in addition to the endogenous Clk.
Expression of ClkT:SV5\V5.WT results in a shortening of the circadian period compared to wild type controls . Flies also exhibit defects in behavioural rhythms seen in wild type flies in response to daily temperature cycles (in the absence of light).
Clkout/ClkTag:V5.WT is a non-enhancer of abnormal locomotor rhythm phenotype of nmoP1/Df(3L)Exel6279
Clkout/ClkTag:V5.WT is a non-suppressor of abnormal locomotor rhythm phenotype of nmoP1/Df(3L)Exel6279
Expression of ClkT:SV5\V5.WT (in a Clkout mutant background) does not alter the locomotor rhythm defects seen in nmoP1/Df(3L)Exel6279.
ClkTag:V5.WT rescues the circadian rhythm defects observed in Clkout homozygous adults.
Expression of ClkT:SV5\V5.WT rescues the behavioral rhythm defects seen in Clkout following following exposure to entrainment conditions (12 hours light: 12 hours dark followed by several days in constant darkness). A similar rescue is seen at 18, 25 and 29[o]C. The failure of Clkout mutant flies to entrain to daily temperature cycles in the absence of light is also rescued in these flies.
Expression of ClkT:SV5\V5.WT rescues the locomotor activity rhythm defects in >90% of Clkout flies with periods of ~24.2 hours.
