UAS regulatory sequences drive expression of a mutant form of Hsap\TARDBP lacking the NLS sequence. Six basic amino acids have been replaced with alanine (K82A, R83A, K84A, K95A, K97A, R98A).
The expression of Hsap\TARDBPNLSmt.Scer\UAS under the control of Scer\GAL4GMR.PU leads to eye morphology defects in 5 days old adults, including a severe decrease in retinal thickness, as compared to controls.
Flies expressing Hsap\TARDBPNLSmt.Scer\UAS under the control of Scer\GAL4GMR.PU exhibit more severe vacuolar retinal degeneration and reduction in retinal thickness compared with wild type Hsap\TARDBP (Hsap\TARDBPScer\UAS.cIa).
