Strong expression of RhoGAP68FScer\UAS.T:Disc\RFP-mCherry under the control of Scer\GAL4Dll-md23 results in pupal lethality and necrosis of the tarsus.
Moderate expression of RhoGAP68FScer\UAS.T:Disc\RFP-mCherry under the control of Scer\GAL4Dll-md23 results in tarsal morphology defects, with a low incidence of broken tarsi. At the prepupal stage, 12% of legs show tears in the epithelium, proximal to the nascent tarsal joints. The tears form along the lateral membrane domain, below the adherens junctions and septate junctions and above the basal surface of the epithelium.
Moderate expression of RhoGAP68FScer\UAS.T:Disc\RFP-mCherry under the control of Scer\GAL4ptc-559.1 alters the morphology and distribution of Rab4-positive endosomes in the leg disc epithelium. They show decreased accumulation in the apical region, increased accumulation in the medial region, enlargement and increased clustering.
