Detailed Description

SH3PX110A/SH3PX110A or SH3PX110A/Df(3L)BSC392 are viable with impaired fertility. SH3PX110A/Df(3L)BSC392 third instar larvae do not have a significantly different number of total boutons, satellite boutons or active zones per bouton at the neuromuscular junction (NMJ; counted at NMJ 4 and 6/7), compared to wild type. Mutants also have similar active zone density and active zone length compared to wild type; synaptic vesicle numbers are also similar to wild type, but SH3PX110A/Df(3L)BSC392 mutants show a small but significant increase in vesicle diameter at synapses and active zone T-bars shape is frequently malformed.

SH3PX110A/Df(3L)BSC392 or SH3PX110A/SH3PX1C1 third instar larvae do not have significantly different miniature excitatory junction potential (mEJP) amplitude, but do have significant reduction in evoked EJP amplitude and quantal content (number of vesicles released per action potential), at the NMJ compared to controls.

SH3PX110A/SH3PX1C1 nwk2/nwk2 double mutants do not have significantly different quantal content deficits or further reduced evoked EJP amplitude compared to nwk2/nwk2 mutants at the third instar larval neuromuscular junction (though there is a trend towards reduced quantal content and EJP amplitude).

Comments

Neuronal (Scer\GAL4elav-C155) but not muscle (Scer\GAL4how-24B) driven expression of SH3PX1Scer\UAS.T:Zzzz\FLAG rescues quantal content and EJP amplitude to wild type levels at the neuromuscular junction of SH3PX110A/SH3PX1C1 third instar larvae.
