Under constant darkness conditions, most flies expressing domHMC04451 under the control of Scer\GAL4tim.PE, alone or in combination of Scer\GAL80Pdf.PS, lose circadian rhythmicity and periods become longer, as compared to controls; this phenotype is not observed when expression is under the control of Scer\GAL4P2.4.Pdf alone. Longer periods are also observed when the Scer\GAL4tim.PE, Scer\GAL80Pdf.PS-regulated expression is restricted to adulthood (by using Gal80[ts] for the temporal control of expression).
The expression of domHMC04451 under the control of Scer\GAL4tim.PE alone blunts the morning activity peak.
The expression of domHMC04451 under the control of Scer\GAL4tim.PE or Scer\GAL4P2.4.Pdf does not lead to obvious defects in the brain structure, but leads to a decrease in the number of lLNv neurons. In the Scer\GAL4P2.4.Pdf settings, the lLNv neurons also have shorter dorsal projections. Expression under the control of Scer\GAL4tim.PE also leads to a decrease in the number of sLNv neurons.
Expression of domHMC04451 under the control of Scer\GAL4Act5C.PI results in pupal lethality.
Expression of domHMC04451 under the control of Scer\GAL4bbg-C96 results in most flies exhibiting defective wing margins, and a small subset showing wing nicks, in contrast to controls.
Expression of domHMC04451 under the control of Scer\GAL4nos.UTR.T:Hsim\VP16, Scer\GAL4nos.PG and Scer\GAL4otu.T:Hsim\VP16 causes significantly decreased female fertility, small ovaries and defects in cyst differentiation in the germarium, as compared to controls.
Expression of domHMC04451 under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 does not affect the development of the germarium or ovariole, but results in a significant proportion of eggs showing the 'dumpless' egg phenotype, as shown by defective late stage egg chambers with short eggs and non-fragmented nurse cells, and a significant reduction in egg-laying, as compared to controls.
Expression of domHMC04451 under the control of Scer\GAL4tj.PU or Scer\GAL4C587 leads to severe packaging defects during oogenesis, ranging from two cysts in a single egg chamber to complete ovariole fusions; many germaria show additional stalk-like structures without germline cysts, and egg laying is nearly completely abolished, as compared to controls.
