Expression of Mmus\BaxScer\UAS.cGb under the control of Scer\GAL4Hml.Δ leads to increased susceptibility to Leishmania infection, with significantly increased lethality upon treatment with Leishmania amastigotes, but no significant difference in survival when untreated, as compared to wild type.

Expression of Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4vg.PM results in an increase in lethality, a notched wing phenotype of variable severity in adults and increased apoptosis in the wing disc in third instar larvae.

Expression of Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4da.G32 results in lethality at embryonic stages.

Expression of Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4Gpb5 eliminates Gpb5/Gpa2 neuroendocrine cells.

Mmus\BaxScer\UAS.cGa expression in the wing (under the control of Scer\GAL4vg.PM) leads to apoptosis in the wing tissue that is not essential to fly survival, thus adult wings exhibit notches on their wing margins. The phenotype is variable, with weak (notches scattered along the margin without significant change in wing size), intermediate (wings are reduced in size and wing margin is lost) and strong (partially or completely absent wings) phenotypes.

Scer\GAL4GMR.PF-mediated expression of Mmus\BaxScer\UAS.cGb results in eye ablation.

Scer\GAL41118-driven Mmus\BaxScer\UAS.cGa expression results in 85% lethality, mostly at embryonic stages, and half of the surviving adults have shrivelled wings. Surviving individuals with expanded or shrivelled wings do not show major differences in their viability, at least up to 10-15 days of age.

No clear rhythm is displayed in average activity plots by the Scer\GAL41118-driven Mmus\BaxScer\UAS.cGa expressing flies in constant darkness. At the individual level, 83% percent of the adult Scer\GAL41118-driven Mmus\BaxScer\UAS.cGa expressing flies display arrhythmic locomotor activity in constant darkness. The remaining rhythmic flies show a shortening of the period with a mean value of about 22.5h and a low power.

In light dark conditions the Scer\GAL41118-driven Mmus\BaxScer\UAS.cGa expressing flies show a shift of evening activity peak of about 2h earlier than the controls.

Ubiquitous expression of Mmus\BaxScer\UAS.cGb under the control of Scer\GAL4da.G32 affects the pattern of programmed cell death through embryonic development. For instance, from stage 11 to 13, the segmental pattern of dying cells along the antero-posterior axis normally found in wild-type embryos is disorganised. Cell aggregates, intensely-stained with Acridine Orange are seen in these embryos, likely as a result of engulfment of the apoptotic bodies by macrophages. The morphology of these embryos is hardly recognizable, and most of them die before hatching. Adults are rarely found.

Mmus\BaxScer\UAS.cGb heterozygotes (i.e. with no Scer\GAL4 driver) are sub-viable. Flies homozygous for Mmus\BaxScer\UAS.cGb have a 2 day lifespan and reduced fertility. These flies also show melanotic masses at larval and adult stages.

Eye-specific expression of Mmus\BaxScer\UAS.cGb (through Scer\GAL4GMR.PF) alters the ordered array of ommatidia and bristles, normally found in the wild-type in 45% of these flies. Many ommatidia are missing in these flies, with those remaining appearing smaller, with loss of pigments cells, photoreceptors and/or cone cells. Holes and vacuoles are also found, indicating massive levels of cell death.

Expression of Mmus\BaxScer\UAS.cGb in the developing wing (through Scer\GAL4vg.PM) reduces the size of the wings in a temperature-dependent manner. At 18[o]C, most of the flies have notched wings, while at 21[o]C, the wing phenotype ranges from notched to absent wing. In some cases, the wing appears as a tubular structure, probably because the ventral and dorsal layers which make up the wing do not merge. At 25[o]C, most of the flies exhibit a gnarled or wrinkled wing phenotype.

In the third instar larval imaginal wing disc, Mmus\BaxScer\UAS.cGb expression under the control of Scer\GAL4vg.PM at 18[o]C induces a modification in the size and shape of the disc. at 21[O]c, the wing part of the disc is reduced in size and highly disorganized.

Over-expression of Uba1 with Uba1UY3010 under the control of Scer\GAL4vg.PM attenuates the notched wing phenotype and lethality observed in flies expressing Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4vg.PM.

Uba1s3484/+ fails to suppress the notched wing phenotype and lethality observed in flies expressing Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4vg.PM.

Co-expression of Uba1EP2375 almost completely suppresses the notched wing phenotype observed in adults, and reduces the number of apoptotic cells in third instar wing discs in flies expressing Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4vg.PM.

Co-expression of Uba1EP2375 partially rescues the lethality seen in flies expressing Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4da.G32.

Rpn10Pw+ partially suppresses the notched wing phenotype observed in flies expressing Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4vg.PM.

The presence of Prosα2EP931, slmb8 or slmb41 enhances the notched wing phenotype observed in flies expressing Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4vg.PM.

Co-expression of SkpAEP1423 partially suppresses the notched wing phenotype observed in flies expressing Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4vg.PM.

The presence of morgueEP1184 does not significantly affect the notched wing phenotype observed in flies expressing Mmus\BaxScer\UAS.cGa under the control of Scer\GAL4vg.PM.

Ablation of hemocytes (through expression of Mmus\BaxScer\UAS.cGb in the hemocytes under the control of Scer\GAL4He.PZ) does not suppress the "black dot" phenotype seen in MAPk-Ak2Δ43 mutant larvae.

A mgeB10 heterozygous background partially suppresses the wing phenotypes found upon expression of Mmus\BaxScer\UAS.cGa in the wing under the control of Scer\GAL4vg.PM. A shift in phenotype distribution from about 60% of strong and about 40% of intermediate phenotypes in the control cross to 30% of strong and about 70% of intermediate phenotypes in these double mutants.

A Tom70KG06223 heterozygous background partially suppresses the wing phenotypes found upon expression of Mmus\BaxScer\UAS.cGa in the wing under the control of Scer\GAL4vg.PM.

A Tom7BG02496 heterozygous background fails to suppresses the wing phenotypes found upon expression of Mmus\BaxScer\UAS.cGa in the wing under the control of Scer\GAL4vg.PM.

A unc-4503692 heterozygous background enhances the wing phenotypes found upon expression of Mmus\BaxScer\UAS.cGa in the wing under the control of Scer\GAL4vg.PM. There is a significant increase from 60% to 90% of strong phenotypes (compared to controls) and a decrease from 40% to 10% of intermediate phenotypes. This shift in phenotype distribution reflects an enhancement of the Mmus\BaxScer\UAS.cGa-induced phenotype thus showing an increase of Mmus\BaxScer\UAS.cGa-induced cell death.

The eye ablation phenotype of Scer\GAL4GMR.PF, Mmus\BaxScer\UAS.cGb is partially restored in a debclKO-unspecified heterozygous background and nearly fully restored in a debclKO-unspecified homozygous background.

Ubiquitous co-expression of Hsap\BCL2Scer\UAS.cGa and Mmus\BaxScer\UAS.cGb under the control of Scer\GAL4GMR.PF results in a pattern of cell death similar to flies expressing Hsap\BCL2Scer\UAS.cGa alone.

Expression of Hsap\BCL2Scer\UAS.cGa is able to rescue the lethality of Mmus\BaxScer\UAS.cGb-expressing flies at 21[o]C (both under the control of Scer\GAL4vg.PM).

Co-expression of Hsap\BCL2Scer\UAS.cGa in Mmus\BaxScer\UAS.cGb-expressing flies at 18[o]C induces a decrease in the ratio of strong wing phenotypes as opposed to intermediate and weak phenotypes and an increase of the ratio of weak wing phenotypes as opposed to intermediate and strong phenotypes. No significant effect was observed at 21 and 25[o]C. The structure of the wing is partially restored in these double mutants.
