No clear rhythm is displayed in average activity plots by the Scer\GAL41118-driven Mmus\BaxScer\UAS.cGa expressing flies in constant darkness. At the individual level, 83% percent of the adult Scer\GAL41118-driven Mmus\BaxScer\UAS.cGa expressing flies display arrhythmic locomotor activity in constant darkness. The remaining rhythmic flies show a shortening of the period with a mean value of about 22.5h and a low power.
In light dark conditions the Scer\GAL41118-driven Mmus\BaxScer\UAS.cGa expressing flies show a shift of evening activity peak of about 2h earlier than the controls.
Ubiquitous expression of Mmus\BaxScer\UAS.cGb under the control of Scer\GAL4da.G32 affects the pattern of programmed cell death through embryonic development. For instance, from stage 11 to 13, the segmental pattern of dying cells along the antero-posterior axis normally found in wild-type embryos is disorganised. Cell aggregates, intensely-stained with Acridine Orange are seen in these embryos, likely as a result of engulfment of the apoptotic bodies by macrophages. The morphology of these embryos is hardly recognizable, and most of them die before hatching. Adults are rarely found.
Mmus\BaxScer\UAS.cGb heterozygotes (i.e. with no Scer\GAL4 driver) are sub-viable. Flies homozygous for Mmus\BaxScer\UAS.cGb have a 2 day lifespan and reduced fertility. These flies also show melanotic masses at larval and adult stages.
Eye-specific expression of Mmus\BaxScer\UAS.cGb (through Scer\GAL4GMR.PF) alters the ordered array of ommatidia and bristles, normally found in the wild-type in 45% of these flies. Many ommatidia are missing in these flies, with those remaining appearing smaller, with loss of pigments cells, photoreceptors and/or cone cells. Holes and vacuoles are also found, indicating massive levels of cell death.
Expression of Mmus\BaxScer\UAS.cGb in the developing wing (through Scer\GAL4vg.PM) reduces the size of the wings in a temperature-dependent manner. At 18[o]C, most of the flies have notched wings, while at 21[o]C, the wing phenotype ranges from notched to absent wing. In some cases, the wing appears as a tubular structure, probably because the ventral and dorsal layers which make up the wing do not merge. At 25[o]C, most of the flies exhibit a gnarled or wrinkled wing phenotype.
In the third instar larval imaginal wing disc, Mmus\BaxScer\UAS.cGb expression under the control of Scer\GAL4vg.PM at 18[o]C induces a modification in the size and shape of the disc. at 21[O]c, the wing part of the disc is reduced in size and highly disorganized.
