Description

Imprecise excision of P{EP}G9172 results in deletion of the Tnks transcription and translation initiation sites (-20 to +1839 with reference to the Tnks transcriptional start site); approximately 3.3K nucleotides from P{EP}G9172 remain at this site.

Detailed Description

Tnks19/Tnks503 transheterozygous mutants eclose at normal rates and are viable, but adults display a significant increase in mortality when maintained on a sucrose only diet (5% sucrose (v/v) in aqueous solution), as compared to controls.

Tnks19/Tnks503 transheterozygous mutant adult midguts show a significant increase in the numbers of intestinal stem cells, of intestinal progenitor cells and of mitotic cells, but similar apoptotic levels, as compared to controls; these phenotypes become more severe with age.

The increase in the numbers of intestinal progenitor cells and mitotic cells present in Tnks19/Tnks503 transheterozygous mutant adult midguts is suppressed by AxnS044230 heterozygosity; the increase in the numbers of intestinal stem cells and mitotic cells observed in Tnks19/Tnks503 transheterozygous mutant adult midguts are suppressed by expressing either upd3KK110348 or upd3GD6811 under the control of Scer\GAL4Myo31DF-NP0001.

Expression of AxnScer\UAS.T:SV5\V5 under the control of Scer\GAL471B in the Tnks19/Tnks503 background leads to morphological wing (notches, loss of wing margin, ectopic bristles) and ventral abdomen (loss of sternite and sternite bristles, reduced size of the sternites that remained, expansion of pleura) defects in adults.

Comments

Expressing TnksScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Myo31DF-NP0001 significantly rescues the increase in the numbers of stem cells and mitotic cells observed in the adult midgut of Tnks19/Tnks503 transheterozygous mutant adults.
