Detailed Description

Duba107 homozygotes are viable and develop normally, they do not show any defects in developmental cell death during either eye (removal of interommatidial cells) or arista (branching pattern) development but homozygote males (not females) are sterile and their seminal vesicles do not contain any mature sperm. Duba107 mutant males show defects in spermatid individualization (the spermatids appear thicker and disorganized and the individualization complex, cystic bulges as well as the waste bags are not detectable) and the activity of the caspases involved in the individualization process is reduced. Strong accumulation of ubiquitylated proteins is observed in the mutant testes.

Genetic Interactions
Statement
Reference

The strongly reduced eye size characteristic for adult flies expressing either rprGMR.PU or hidGMR.PG is partially reverted by combination with Duba107.

The spermatogenesis defects characteristic for Duba107 homozygote adult males are not suppressed by expression of any of the following: Droncpro.C-A.Scer\UAS, DriceScer\UAS.T:SV5\V5 or DriceC-A.Scer\UAS.T:SV5\V5 under the control of Scer\GAL4Hsp83.PA in the mutant background as mature spermatids are completely missing in these flies.

Comments

The spermatogenesis defects and male sterility characteristic for Duba107 homozygote males is rescued by expression of DubaScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4bam.T:Hsim\VP16 in the mutant background, whereas expression of either DubaC227A.Scer\UAS.T:Ivir\HA1 or DubaS183A.Scer\UAS.T:Ivir\HA1 fails to rescue the mutant phenotype - on the contrary, the spermatid structures seem to be disrupted even more.
