ScerGAL40098-G4 drives expression in 32 neurons in the brain, including adult subesophageal zone DSOG1 and DSOG2 neurons, 6 neurons in the thoracico-abdominal ganglion, and approximately 10 peripheral neurons that project to the abdominal ganglion.
Inhibition (using Hsap\KCNJ2UAS.EGFP in combination with a Gal80[ts] transgene to restrict expression to the 48 hours of the adult stage prior to experimentation) of Scer\GAL40098-G4 cells significantly and substantially increases water intake of adult females under water-replete conditions, compared to controls; this inhibition causes water-deprived flies to drink to the extent that they become extremely bloated and regurgitate when provided with water, unlike controls. Significant water overconsumption is also observed when this adult-stage inhibition is restricted to a smaller set of cells using Scer\FLP1276B and Scer\GAL80αTub84B.PG, but not Scer\FLP1934 and Scer\GAL80αTub84B.PG; with inhibition of the Scer\FLP1276B subset of Scer\GAL40098-G4 cells also resulting in significant overconsumption of sugar, salt and bitter solutions, as well as absolute ethanol (leading to death), compared to controls; there are significantly more attempts to consume crystallized sucrose and significantly increased proboscis extension probability in response to tarsal water, sugar solutions and bitter solutions (except for where control response is already 100 percent); there is no effect on locomotion under fed or starved conditions, compared to controls.
Inhibition (using Hsap\KCNJ2UAS.EGFP) of the combined set of the Scer\FLP1276B subset (with excised Scer\GAL80αTub84B.PG) of Scer\GAL40098-G4 cells and the Scer\GAL4VT005008 cells, in combination with a Gal80[ts] transgene to restrict expression to the adult stage, in females does not significantly affect proboscis extension in response to bitter solution, but does significantly increase bitter solution consumption, compared to controls, to a similar extent as inactivation of Scer\FLP1276B-expressing Scer\GAL40098-G4 cells alone.
Inhibition (using Hsap\KCNJ2UAS.EGFP) of the combined set of the Scer\FLP1276B subset (with excised Scer\GAL80αTub84B.PG) of Scer\GAL40098-G4 cells and the Scer\GAL4NP0534 cells, in combination with a Gal80[ts] transgene to restrict expression to the adult stage, in females significantly increases proboscis extension in response to bitter solution and significantly increases bitter solution consumption, compared to controls, but overconsumption is significantly reduced compared to inactivation of Scer\FLP1276B-expressing Scer\GAL40098-G4 cells alone.
Acute inhibition (using shi1.UAS at 30[o]C) of the Scer\FLP1276B subset (with excised Scer\GAL80αTub84B.PG) of Scer\GAL40098-G4 cells in fed adult females significantly increases consumption of a bitter solution, compared to controls.
Inhibition (using Ctet\TeTxLCTNT.UAS) of Scer\GAL40098-G4 cells in adult females results in water overconsumption to the extent that the abdomen becomes distended, but does not affect the firing rate of adult subesophageal zone DSOG1 neurons in either fed or water overconsumption conditions.
Thermogenetic activation (using TrpA1UAS.(B).cKa at 30[o]C or higher) of Scer\GAL40098-G4 cells does not affect water consumption of fed adult females, compared to controls. Activation of the Scer\FLP1276B subset (with excised Scer\GAL80αTub84B.PG) of Scer\GAL40098-G4 cells in adult females does not affect consumption of water, sugar solution or bitter solution under water-replete conditions, compared to controls; activation significantly reduces consumption of sucrose solution by starved, water-deprived females and consumption of water by water-deprived females, compared to controls.
