The lesion in the RdlMD-RR allele has been described as A302S in print, but the amino acid replacement is actually A301S.

Mutant larvae are more resistant to fipronil and to pyriprole compared to wild type.

Mutant flies show more than 45,000-fold resistance to dieldrin compared to wild type.

Mutant flies are significantly slower to recover from heat shock compared to wild type.

RdlMD-RR mutants show behavioral convulsions after brief exposure to elevated temperature.

RdlMD-RR neurons have a marked reduction in sensitivity to the picrotoxin blockade of spontaneous GABAergic postsynaptic currents compared to wild type. Additionally, there is a much lower increase in neuron excitability in response to picrotoxin in the RdlMD-RR mutants compared to wild type.

Mutant flies are resistant to dieldrin (more than 55-fold resistant) and α-thujone (5-fold resistant) compared to wild-type flies.

Resistant to cyclodiene. Reduces the single channel conductance by 5% for inward current and 17% for outward current. Single channel current is approximately 60% lower for outward current than for inward current in both wild type and mutant. Increases open time and reduces closed times, reflecting a net stabilization of the channel open state by a factor of approximately five. Shows a marked reduction in the rate of GABA-induced desensitization, and a net destabilization of the desensitized conformation by a factor of 29.

Confers resistance to PTX and cyclodienes.

Resistant to dieldrin selection.

Flies are insensitive to 10μM dieldrin, and less sensitive to picrotoxinin than wild-type.

Flies show approximately 4000-fold resistance to dieldrin compared to wild-type.

The A302S substitution is found in 14 resistant strains of D.melanogaster. The M361I amino acid replacement is found in some, but not all, resistant strains tested so is likely to be a polymorphism unrelated to insecticide resistance.
