Codons D301 and P303 of mAChR-A were replaced by K codons (AAG) via CRISPR. Silent point mutations were also introduced in the PAM region of the guide RNA targets.
Unlike controls, in mAChR-AKK antennal lobe inhibitory local neurons (ILNs), muscarine-evoked currents are not voltage-dependent; unlike controls, a depolarizing pre-pulse step (40 mV, 30 sec) does not potentiated the response to muscarine. ILNs show voltage-independent post-tetanic potentiation and abolished nicotine-induced potentiation. Unlike controls, ILNs do not show a decrease in firing rate upon exposure to the odor (ethyl butyrate); adult show significantly stronger habituation to ethyl butyrate and apple cider vinegar in olfactory avoidance/attraction assays.
