UASt regulatory sequences drive expression of a codon-optimized version of Elp3 that has been engineered (via numerous silent mutations) to be resistant to RNAi mediated by P{NIG.15433R}. Amino acid substitutions Y531A and Y532A have been introduced into the coding sequence (the equivalent residues have previously been shown to be critical for the acetylase and methylase enzymatic activities of the yeast Elp3 ortholog).
