Gene Snapshot
ringmaker (ringer) encodes a protein involved in microtubule bundle formation and polymerization. [Date last reviewed: 2019-09-19]
Protein Function (UniProtKB)
Regulator of microtubules required for axonal extension during embryonic development and axon regeneration following injury (PubMed:27422099, PubMed:31919191). Promotes microtubule bundling and polymerization (PubMed:27422099). Together with futsch, required for neuromuscular junction (NMJ) bouton growth by regulating synaptic microtubules (PubMed:31156389). Function with futsch in maintaining microtubule stability and dynamics, is essential for promoting axon regeneration in response to peripheral (PNS) and central nervous system (CNS) injury (PubMed:31919191). In response to axotomy, acts downstream of a stress response cascade involving Xbp1 splicing, to control axon regeneration (PubMed:31919191).
(UniProt, Q9VV43)
Summary (Interactive Fly)

PNS, maintains microtubule stability/dynamics with the microtubule-associated protein Futsch - splice factor Rtca suppresses Xbp1 via nonconventional mRNA splicing, which in turn reduces ringer expression to inhibit axon regeneration - lies downstream from and is negatively regulated by the microtubule-associated deacetylase HDAC6

Model Confidence:
  • Very high (pLDDT > 90)
  • Confident (90 > pLDDT > 70)
  • Low (70 > pLDDT > 50)
  • Very low (pLDDT < 50)

AlphaFold produces a per-residue confidence score (pLDDT) between 0 and 100. Some regions with low pLDDT may be unstructured in isolation.

Additional Descriptive Data

ringer protein is prominantly expressed in the embryonic ventral midline with some expression in the surrounding CNS and PNS. It is expressed in both neurons and glial cells in the midline. The neurons have been identified as the MP1 neuron starting at embryonic stage 13 and aCC, pCC, and RP2 neurons starting at stage 14.
