Expression of SerBd.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ey.PH results in the loss of the ventral-half of the eye, both at the eye disc and the adult eye level.

Expression of SerBd.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4bi-omb-Gal4 results in suppression of eye development on both the ventral and dorsal margins in the eye disc.

Expression of SerBd.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ey.PH during 12-72 hours of development (using a shift to 29oC from 18oC) generally results in complete elimination of the eye field in the imaginal disc, although antennal development is not affected in the disc. Some extremely small eye discs with a few photoreceptors are also seen. Expression of SerBd.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ey.PH during 72-96 hours of development (using a shift to 29oC from 18oC) shows a significant reduction in the no-eye phenotype along with an increase in frequency of selective suppression of the ventral eye compared to expression during 12-72 hours of development. Expression of SerBd.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ey.PH during 96-168 hours of development (using a shift to 29oC from 18oC) shows a sharp increase in the selective loss of ventral eye phenotype.

Eye discs that contain clones expressing SerBd.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4Act5C.PP are variably reduced. In the most severe cases this results in rudimentary eyes. In some cases a preferential reduction of the ventral eye is seen. Ventral clones in the eye disc expressing SerBd.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4Act5C.PP are only rarely recovered.

When expressed via Scer\GAL4e22c, animals die at approximately the first larval instar with head and mouth hook abnormalities remarkably similar to those of Serrev6-1. When expressed via Scer\GAL4Ser.PGF or Scer\GAL4Ser.PGF flies exhibit wing margin loss phenotypes. The severity of the phenotype is greater for Scer\GAL4Ser.PGF than for Scer\GAL4Ser.PGF. The wing margin phenotype for SerBd.Scer\UAS.T:Hsap\MYC, Scer\GAL4Ser.PGF is more severe in the presence of two wild type Ser alleles than is the wing phenotype associated with the SerBd-3 heterozygous for a single Ser+ allele. When expressed via Scer\GAL4Ser.PGF in a Ser+r83k background the flies have phenotypes indistinguishable from SerBd-3/Ser+r83k mutants. When expressed via Scer\GAL4hs.PB in embryos, lethality results, with neuronal hypertrophy at the expense of hypodermal differentiation. When expressed via Scer\GAL4Ser.PGF, the regions of the wing imaginal disc corresponding to the presumptive wing blade are consistently smaller and misshapen relative to wild type. When expressed via Scer\GAL4ptc-559.1, flies die during early pupal formation prior to imaginal disc eversion.

Expression of SerBd.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ey.PH at 25oC in eye discs homozygous for pnrVX6 (generated using the "EGUF" method to remove all eye disc cells except the homozygous pnrVX6 clone cells) results in a complete loss of the eye in 99% of flies, compared to 50% of flies showing complete loss of the eye or a very small eye when SerBd.Scer\UAS.T:Hsap\MYC is expressed under the control of Scer\GAL4ey.PH at 25oC in a wild-type background. Expression of SerBd.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4ey.PH at 25oC in eye discs homozygous for Df(3L)iro-DFM3 (generated using the "EGUF" method to remove all eye disc cells except the homozygous Df(3L)iro-DFM3 clone cells) results in a complete loss of the eye in 99% of flies, compared to 50% of flies showing complete loss of the eye or a very small eye when SerBd.Scer\UAS.T:Hsap\MYC is expressed under the control of Scer\GAL4ey.PH at 25oC in a wild-type background.

When expressed via Scer\GAL4hs.PB in a Df(3R)Dl-BX6 background, no reduction in the severity of the neurogenic phenotype is observed. Coexpression of DlScer\UAS.cJa and SerBd.Scer\UAS.T:Hsap\MYC via Scer\GAL4Ser.PGF did not alter the extent of cell proliferation in the dorsal wing compartment in discs or adult wing blades. Coexpression of DlScer\UAS.cJa and SerBd.Scer\UAS.T:Hsap\MYC via Scer\GAL4ptc-559.1 leads to variable but consistent reduction in the extent of ventral compartment cell proliferation as compared to that produced by DlScer\UAS.cJa alone. The antimorphic effects of SerBd.Scer\UAS.T:Hsap\MYC (therefore SerBd-3) are compartmentally restricted in the wing disc. Coexpression of SerBd.Scer\UAS.T:Hsap\MYC and fngScer\UAS.cKa via Scer\GAL4hs.PB results in animals with apparently normal CNS development, and apparently improved viability when compared to SerBd.Scer\UAS.T:Hsap\MYC, Scer\GAL4hs.PB alone. When co-expressed with fngScer\UAS.cKa via Scer\GAL4ptc-559.1, flies survive longer, to late pupal stages, after disc eversion and as pharate adults. The flies cannot eclose but exhibit complete though shortened legs and no wing blades.
