Description

Nucleotide 544 (G) has been replaced with CAC, which produces a frameshift after amino acid residue 103 (the protein produced contains 33 novel amino acid residues, which are PLRLPVTRIRPPEAEVAPQQRHPLLCIRQPPRP).

Comment:

The G in codon 104 of mira has been replaced with CAC leading to a frameshift and early translation termination after the addition of 33 novel amino acids. The last wild type amino acid is F103.

Detailed Description

miraL44 mutants exhibit ectopic gcm expression and an increase in glial number (4-5) accompanied by the lack of the neuronal progeny.

All progeny of NB6-4T adopt a glial cell fate, expressing gcm and repo and moving towards the midline.

Axon tracts of embryos are grossly defective, longitudinal connectives are generally broken or severely reduced in width and commissures are partially or completely fused.

Over-expression of CycEScer\UAS.cBa under the control of Scer\GAL4sca-109-68 in a miraL44 background is sufficient to confer neuronal fate to the NB6-4t lineage.

The ovarian phenotype of cup15 and cup21 homozygous females is enhanced if they are also heterozygous for miraL44; a greater fraction of egg chambers arrest development between stages 3 and 5 in the double mutant ovaries compared to in the single mutant ovaries.
