GAL4::VP16 driver sequence (first 147 amino acids of Scer\GAL4, including the DNA binding domain, and the carboxy-terminal 78 amino acids of Hsim\VP16, including the activation domain) is integrated in frame into the N coding region between the TM domain and the NLS1 domain. The fusion protein is expressed from a heat shock promoter.

Expression of N3.hs.T:Scer\GAL4,T:Hsim\VP16 using heat shock (either 1 or 2 hours) during the third instar larval stage enhances the rough eye phenotype of RalaEE1/Y.
The heat-shocked progeny from heterozygous amx1 females crossed to N3.hs.T:Scer\GAL4,T:Hsim\VP16 males hatch at a similar rate to wild type and are viable.
The neurogenic phenotypes seen in stage 8 embryos when homozygous amx1 mutant females are crossed to N3.hs.T:Scer\GAL4,T:Hsim\VP16 males (heat-shocked before the first delamination round) are similar to those seen when amx1 females are crossed to wild type males.
Expression of N3.hs.T:Scer\GAL4,T:Hsim\VP16 using three 40 minute heat shocks starting at 8-9 hours after egg laying does not rescue hub cell number in the gonads of male N264-39 larvae. However, expression of N3.hs.T:Scer\GAL4,T:Hsim\VP16 using three 40 minute heat shocks starting at 5-6 hours after egg laying results in significant rescue of hub cell number in the gonads of male N264-39 larvae, with almost half of the animals reaching the wild-type range of hub cells.
