Description

Insertion of a P{EP} element 235bp upstream of the 5' end of exon 1a. The P{EP} element is inserted so that a sense Bx transcript is produced by transcription from the promoter in the P{EP} element.

Insertion of a P{EP} element upstream of the 5' end of the Bxa transcript.

Mutant flies show increased sensitivity to ethanol sedation compared to control flies.

Expression of BxEP1306 under the control of Scer\GAL4Ccap.PP gives rise to flies that fail to expand their wings after eclosion.

Mis-expression of BxEP1306 under the control of Scer\GAL4Ccap.PP for the entire pupal period (using Scer\GAL80ts.αTub84B at 25[o]C to block expression up to the pupal period) leads to a fully penetrant wing expansion defect. Mis-expression 40 hours after pupal formation, however, does not affect post-eclosion behavior.

Mutant animals exhibit an increased sensitivity to cocaine. Mutants exposed to 75ug of cocaine behave similarly to control flies exposed to 100ug of the drug. The number and detailed morphology of LNv neurons is normal in mutant animals. Mutant adults exhibit a less robust locomotor rhythm in constant darkness than control flies.

When clones are made in the wing disc that express BxEP1306 (driven by Scer\GAL4Act5C.PP), they are round in shape with smooth borders and sort into the ventral compartment. When clones are made in the wing disc expressing BxEP1306, they do not respect the dorsal ventral boundary.

Expression of BxEP1306 under the control of Scer\GAL4sca-537.4 results in the loss of several scutellar and dorsocentral macrochaetae.

Flies expressing BxEP1306 under the control of Scer\GAL4ap-md544 show loss of the normal wing margin and sporadic patches of ectopic wing margin, which promote local overgrowth.

Flies expressing BxEP1306 under the control of Scer\GAL4bi-omb-Gal4 lack most of the normal wing margin and have irregular pieces of ectopic margin associated with overgrowths in the dorsal surface of the wing.

Flies expressing BxEP1306 under the control of Scer\GAL4ptc-559.1 show ectopic wing formation on the wing or notum.

Expression of ChiFL.Scer\UAS.T:Hsap\MYC with BxEP1306, both under the control of Scer\GAL4Ccap.PP, suppresses the wing expansion defect found upon expression of BxEP1306 alone.

Expression of tupScer\UAS.cTa with BxEP1306, both under the control of Scer\GAL4Ccap.PP, suppresses the wing expansion defect found upon expression of BxEP1306 alone.

Expression of dimmScer\UAS.T:Hsap\MYC with BxEP1306, both under the control of Scer\GAL4Ccap.PP, fails to suppress the wing expansion defect found upon expression of BxEP1306 alone.

Expression of Lim3Scer\UAS.cTa with BxEP1306, both under the control of Scer\GAL4Ccap.PP, fails to suppress the wing expansion defect found upon expression of BxEP1306 alone.

When clones are made expressing both BxEP1306 and NECN.Scer\UAS.cGa or fngScer\UAS.B.T:Hsap\MYC are made some of them are round in shape, others more irregular and do not sort into the ventral compartment.

The phenotype of flies expressing BxEP1306 under the control of Scer\GAL4ap-md544 is not suppressed by coexpression of apScer\UAS.cMa. Flies coexpressing BxEP1306 and ap::ChiChAp.Scer\UAS under the control of Scer\GAL4ap-md544 have a normal wing margin and resemble the Bx- mutant wing.

Selected as: a P{EP} insertion line that modifies the pnrMD237/+ phenotype when expressed using Scer\GAL4pnr-MD237.

Comments
Comments

The cocaine sensitivity phenotype is revertable by excision of the P{EP} insertion.
