Flies expressing sggEP1576 under the control of Scer\GAL4P0.5.Pdf (limited to the adult stages using Scer\GAL80ts.αTub84B) have a short period behavioural rhythm of 21.4 hours in complete darkness (DD).

Approximately 50% of flies expressing sggEP1576 under the control of Scer\GAL4tim.PE show rhythmic locomotor activity under constant light conditions, in contrast to wild-type flies, which are uniformly arrhythmic under these conditions.

Flies expressing sggEP1576 under the control of Scer\GAL4tim.PE in a Scer\GAL80Pdf.PS background show rhythmic locomotor activity under constant light conditions.

Flies expressing sggEP1576 under the control of Scer\GAL4tim.PE in a Scer\GAL80cry.PS background are completely arrhythmic for locomotor activity under constant light conditions, as are wild-type flies.

Flies expressing sggEP1576 under the control of Scer\GAL4tim.PE have a locomotor activity period of 20.3 hours under constant darkness conditions.

Flies expressing sggEP1576 under the control of Scer\GAL4P2.4.Pdf are completely arrhythmic for locomotor activity under constant light conditions, as are wild-type flies.

The phase response curve of flies expressing sggEP1576 under the control of Scer\GAL4P2.4.Pdf is indistinguishable from that of controls.

The phase response curve of flies expressing sggEP1576 under the control of Scer\GAL4tim.PE in either a wild-type or Scer\GAL80Pdf.PS background is altered compared to wild-type, having attenuated delay zone amplitudes and earlier transitions (breakpoints) between the delay and advance zones.

Flies expressing sggEP1576 under the control of Scer\GAL4P2.4.Pdf show robust advancement of morning anticipation under standard light-dark conditions, but show no significant change in the phase of the evening output. If these flies are entrained to a short photoperiod regime (winter-like; light:dark 10:14 hours), the evening and morning activities are advanced. If these flies are entrained to a long photoperiod regime (summer-like; light:dark 14:10 hours), principally the morning activity is enhanced.

Flies expressing sggEP1576 under the control of Scer\GAL4tim.PE in either a wild-type or Scer\GAL80Pdf.PS background show robust advancement of morning anticipation under standard light-dark conditions and the evening activity is also advanced. If these flies are entrained to a short photoperiod regime (winter-like; light:dark 10:14 hours), only the evening activity is advanced. If these flies are entrained to a long photoperiod regime (summer-like; light:dark 14:10 hours), both the evening and morning activities are advanced.

sggEP1576 flies show wild-type olfactory habituation.

Expression of sggEP1576 under the control of Scer\GAL4ey.PH results in a normal looking eye.

sggEP1576, Scer\GAL4tim.PE mutant flies in constant darkness (DD) exhibit a period 3h shorter than control flies. Both the average length of subjective day and the period are reduced. sggEP1576, Scer\GAL4P2.4.Pdf mutant flies in constant darkness (DD) exhibit a period a couple of hours shorter than control flies. The period of evening activity is significantly shorter than controls in these flies. sggEP1576, Scer\GAL4cry.PE mutant flies in constant darkness (DD) has no effect on circadian rhythms.

Only 57% of adults expressing sggEP1576 under the control of Scer\GAL4tim.PE have show rhythmic locomotor activity under free-running conditions. Those animals that are rhythmic have a shorter locomotor activity period than normal.

When sggEP1576 is driven by Scer\GAL4tim.Scer\UAS or Scer\GAL4tim.PE the locomotor activity period is decreased by approximately 3 hours.

Expression of Rho1Scer\UAS.cMa suppresses the shortening of the behavioural rhythm period seen when sggEP1576 is expressed under the control of Scer\GAL4P0.5.Pdf (limited to the adult stages using Scer\GAL80ts.αTub84B)in complete darkness (DD).

Flies expressing sggEP1576 under the control of Scer\GAL4tim.PE in a cryb background have a locomotor activity period of 22.6 hours under constant darkness conditions, which is longer (and closer to that of wild type) than the locomotor activity period of 20.3 hours seen under these conditions in flies expressing sggEP1576 under the control of Scer\GAL4tim.PE in wild-type background.

sggEP1576, Scer\GAL4tim.PE, Scer\GAL80Pdf.PS mutant flies in constant darkness (DD) exhibit an evening phase similar to sggEP1576, Scer\GAL4tim.PE animals but have a period of 23.8 hours. The length of subjective day for these animals is about 10.35h. sggEP1576, Scer\GAL4tim.PE, Scer\GAL80cry.PS mutant flies in constant darkness (DD) exhibit an evening phase similar to controls.

More than 80% of adults expressing sggEP1576 under the control of Scer\GAL4tim.PE in a timblind background rapidly become arrhythmic for locomotor activity after they are placed in constant darkness conditions. The few animals that are rhythmic have a locomotor activity period that is intermediate between that of timblind homozygotes and that of adults expressing sggEP1576 under the control of Scer\GAL4tim.PE in a wild-type background.

Excision of the P{EP}sggEP1576 insertion reverts the phenotype to wild-type.

With respect to circadian period length, the following alleles can be ranked from longest to shortest: sggEP1576 = sggtwkr/+ = sgg1/sggEP1576 < sggtwkr/sggtwkr < sgg1/sggtwkr.
