Expressing Rho1Scer\UAS.cMa for 12 hours under the control of Scer\GAL4P0.5.Pdf (restricted to the adult stages using Scer\GAL80ts.αTub84B) significantly reduces the spread of s-LNv neuron projections usually seen at ZT24 (dawn). This dusk-like retracted state is still observed 2.5 days after induction. When Rho1Scer\UAS.cMa is expressed for 12 hours starting at ZT24 (dawn), a normal spread of projections is seen at ZT12 (dusk).

Expression of Rho1Scer\UAS.cMa under the control of Scer\GAL4P0.5.Pdf (restricted to the adult stages using Scer\GAL80ts.αTub84B) suppresses the activity-dependent spreading of s-LNVs seen in controls. In controls, three hours of neuron activation beginning when projections are retracted at dusk (ZT12) results in increases in spread and axonal volume similar to those normally seen at dawn (ZT24). This increase in spread is not observed upon co-expression of Rho1Scer\UAS.cMa. The usual increase in clock protein levels seen upon LNv firing is also disrupted by Rho1Scer\UAS.cMa.

Flies expressing Rho1Scer\UAS.cMa under the control of Scer\GAL4P0.5.Pdf (restricted to the adult stages using Scer\GAL80ts.αTub84B) are either arrhythmic or weakly rhythmic in complete darkness (DD). The phase of clock proteins in DN1 clock neurons is shifted by ~6 hours between control and Rho1Scer\UAS.cMa-induced flies after three days in DD.

In contrast to controls, which adjust their morning and evening activity peaks to align with seasonal dawn and dusk, flies expressing Rho1Scer\UAS.cMa under the control of Scer\GAL4P0.5.Pdf (restricted to the adult stages using Scer\GAL80ts.αTub84B) are active prior to dawn when exposed to a winter light:dark cycle (10 hours day:14 hours night). Timing of activity is unaffected compared to controls when the flies are exposed to a summer cycle (14 hours day:10 hours night).

Expressing Rho1Scer\UAS.cMa for 14 hours under the control of Scer\GAL4P0.5.Pdf (restricted to the adult stages using Scer\GAL80ts.αTub84B) reduces the number of active zones in LNv projections at ZT2 to levels normally seen in controls at ZT14. The normally low number of active zones seen at ZT14 in controls are unaffected by Rho1Scer\UAS.cMa.

Expression of Rho1Scer\UAS.cMa in a stripe of cells at the anterior/posterior boundary of the larval wing disc under the control of Scer\GAL4ptc-559.1 results in cell migration.

Scer\GAL4GMR.PF-mediated expression of Rho1Scer\UAS.cMa results in a rough eye.

Expression of Rho1Scer\UAS.cMa under the control of Scer\GAL4dpp.blk1 induces ectopic apoptosis in third instar wing imaginal disc cells.

Expression of Rho1Scer\UAS.cMa under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 does not block germ cell transepithelial migration or later germ cell migration.

In Rho1Scer\UAS.cMa; Scer\GAL4ptc-559.1 larvae, cells immediately anterior to the anterior-compartment boundary lose their apical profiles, delaminate from the epithelium and show increased levels of apoptosis.

When Rho1Scer\UAS.cMa is driven by Scer\GAL4GMR.PF a rough eye phenotype is seen.

Expression of Rho1Scer\UAS.cMa under the control of Scer\GAL4OK107 does not result in axon growth defects in the mushroom body.

Expression of Rho1Scer\UAS.cMa by Scer\GAL4nos.UTR.T:Hsim\VP16 has no effect on germ cell migration.

Cells expressing Rho1Scer\UAS.cMa under the control of Scer\GAL4Bx-MS1096 in the wing disc mislocalise F-actin and lose epithelial characteristics. Cells lacking apical-basal polarity accumulate basal to the epithelial layer in these discs. Cells in the wing disc marked by Avic\GFP expression and expressing Rho1Scer\UAS.cMa under the control of Scer\GAL4ptc-559.1 disperse from their normal position and can invade adjacent regions of the disc.

Rho1Scer\UAS.cMa driven by Scer\GAL4prd.RG1 has no effect on cytokinesis or mitosis.

Comments

Rho172O clones are rescued by expression of Rho1Scer\UAS.cMa. In some cases, a decrease in the apical area is observed, perhaps due to the high level of Rho1Scer\UAS.cMa overexpression.

Rho172F clones are rescued by expression of Rho1Scer\UAS.cMa. In some cases, a decrease in the apical area is observed, perhaps due to the high level of Rho1Scer\UAS.cMa overexpression.

Expression of Rho1Scer\UAS.cMa specifically in the hemocytes (using the Scer\GAL4crq.PA driver) resuces the hemocyte recruitment defect seen in Rho11B mutants.
