Homozygous Cap-D3EY00456 and Cap-D3EY00456/Df(2L)Exel6023 mutant males are completely sterile when mated to wild-type females.
In male Cap-D3EY00456 mutants, chromosomal organizational steps throughout prophase I are defective, as normal territory formation is never observed in 100% of S2, S4, and S6 stages. Instead, chromatin is seemingly dispersed within the nucleus.
In two cysts observed from Cap-D3EY00456 homozygous males, 7 of 20 anaphase I figures are bridged. This anaphase I bridging most likely represents a failure to resolve chromosomal associations prior to segregation as chromatin appears to be stretched between chromosomes moving to opposing poles.
Polyteny persists in 91.3% of the nurse cells in stage 10 egg chambers of Cap-D3EY00456/Df(2L)Exel7023 females (in contrast to wild-type nurse cells, which disassemble their polytene chromosomes during mid-oogenesis).

The polytene chromosome disassembly that is seen in larval salivary glands expressing Cap-H2EY09979 under the control of Scer\GAL4hs.PB is suppressed by Cap-D3EY00456/Df(2L)Exel7023.
