In Pxtf01000/Pxtf01000 females, both anterior and posterior nurse cells have a greatly enlarged and fragmented nucleolus lacking interconnected tubules during oogenesis, with a greater severity at stage 10B than at stages 8, 9 and 10A. Additionally, their inability to form cytoplasmic actin bundles and to complete dumping by stage 13, results in larger nurse cells and a smaller oocyte than controls. Nurse cells further exhibit an excess of lipid droplets in the cytoplasm during oogenesis.

Homozygous pxtf01000 females are sterile.

pxtf01000 mutant follicles frequently show defects in nurse cell dumping.

pxtf01000 mutant ovarioles typically contain fewer egg chambers than wild-type ones. These mutant ovarioles are deficient in maturing follicles. Approximately 25% of pxtf01000 ovarioles contain a fusion of two adjacent egg chambers and in about half of the ovarioles there is at least one degenerating follicle.

pxtf01000 nurse cell plasma membranes are unstable and frequently break down to produce multinucleate nurse cells in which an actin-rich aggregate containing ring canal remnants remains. Membrane breakdown preferentially occurs in anterior nurse cells. The membrane breakdown, fusion and degeneration defects increase with age, because in 25-day-old pxtf01000 females, the ovarioles are reduced to a single fused structure.

In both homozygotes and trans-heterozygotes of pxtf01000 and pxt[EY03052], more than 50% of stage 9-10 border cell clusters do not complete migration completely.
