Drives expression in neurons of the fan-shaped body and mushroom body, including the mushroom body pedunculus-medial lobe arborizing neuron 1 and MB-AMP neuron of the dopaminergic PPL1 cluster
ScerGAL4c061 drives expression in two MB-MP1 and one MB-AMP neurons of the dopaminergic PPL1 cluster.
Using GFP reconstitution across synaptic partners (GRASP) in MB-MP1 dopaminergic neurons labelled by ScerGAL4c061 and octopaminergic neurons labelled by ScerGAL4Tdc2.PC, reveals expression of GFP in the anterior medial protocerebrum and in the mushroom body heel.
ScerGAL4c061 drives expression in neurons of the dorsal protocerebrum, mushroom body and fan-shaped body.
Inhibition (using shi1.UAS at 33[o]C) of a subset of Scer\GAL4c061 cells (restricted using Scer\GAL80Mef2.mb247) in adults 30 min before and during testing, does not affect 3 h appetitive olfactory memory in starved adults, but does significantly increase memory in flies that are fed between training and testing, compared to controls.
Inhibition of neural activity in Scer\GAL4c061-expressing neurons does not affect sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503). As in wild type flies, courtship defects are seen in response to (S,Z,Z)-CH503.
Thermogenetic activation (using TrpA1UAS.(B).cKa at 30[o]C) of a subset of Scer\GAL4c061 cells in adults (activation restricted using Scer\GAL80Mef2.mb247), in combination with a paired odor, results in a robust increase in 2-min aversive olfactory memory, compared to controls, and this persists for 2 hours; the increase in 2-min memory is significantly reduced when activation is restricted to a smaller subset of Scer\GAL4c061 cells using Scer\GAL80ple.PK or Scer\GAL80ChAT.3.3P.
