Drives expression in neurons projecting to fan-shaped body layer 5.

ScerGAL4c205 drives expression in layer 5 of the fan-shaped body, co-localising with Mip protein.

Expression of ScerGAL4c205 was seen in eight to ten fan-shaped F5 neurons with somata that are loosely arranged near the calyx of the mushroom body at each side of the protocerebrum. The primary neurites of these cells converge into a bundle that runs ventrally and anteriorly into the superior medial protocerebrum.

    Detailed Description

Acute inhibition (using shi1.UAS at 30[o]C) of a subset of Scer\GAL4c205 cells (expression restricted using Scer\GAL80Mef2.mb247) in adults does not affect electric shock avoidance or avoidance of higher temperatures, compared to controls, and there is no effect on locomotion activity index; inhibition of this subset during testing alone, but not training alone, significantly reduces short-term aversive, but not appetitive, olfactory memory, compared to controls.

Inhibition (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4c205 cells, in combination with a Gal80[ts] transgene to begin expression three days prior to experiments, in adults does not affect electric shock avoidance, compared to controls, and there is no effect on locomotion activity index.

Optogenetic activation (using Crei\ChR1::Vcar\ChR1::Vcar\ChR2ReaChR.UAS.Citrine with (orange) light) of Scer\GAL4c205 cells in one chamber of a T-maze does not significantly increase in avoidance of that chamber, compared to controls, and there is no effect on locomotion activity index.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 26[o]C) of a subset of Scer\GAL4c205 cells (expression restricted using Scer\GAL80Mef2.mb247) in adults has no effect on locomotion activity index, compared to controls.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 26[o]C) of a subset of Scer\GAL4c205 cells (expression restricted using Scer\GAL80Mef2.mb247) in adults, in combination with a paired odor, does not affect aversive olfactory memory, compared to controls, and there is no effect on locomotion activity index.

Inhibition of neural activity in Scer\GAL4c205-expressing neurons does not affect sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503). As in wild type flies, courtship defects are seen in response to (S,Z,Z)-CH503.

Activation of fan-shaped body dopaminergic neurons through overexpression of temperature sensitive TrpA1 under the control of Scer\GAL4c205 produces a large increase in the amount of sleep seen in mature (7-8 day old) flies, but has a less severe effect on young (1 day old) flies.

Inhibition (using shi1.UAS at 31[o]C) of Scer\GAL4c205 cells in adults significantly reduces short-term aversive visual, but not olfactory, unimodal memory, compared to controls; inhibition does not affect short-term aversive memory for tested olfactory cues that are associated with conditioned visual cues, or for tested visual cues that are associated with conditioned olfactory cues, compared to controls.

Inhibition (using shi1.UAS at 31[o]C) of the combined set of Scer\GAL4Mef2.247 cells and Scer\GAL4c205 cells in adults significantly reduces short-term aversive memory for tested visual cues that are associated with conditioned olfactory cues, and for tested olfactory cues that are associated with conditioned visual cues, compared to controls.
