The expression of LamCUAS.cSa under the control of Scer\GAL4fln.IFM, Scer\GAL4Act88F.PB or Scer\GAL4EDTP-DJ694 does not affect flight ability; this occurs in an otherwise wild-type background or in a LamCEX296 heterozygous background.

Expressing LamCUAS.cSa under the control of Scer\GAL4Act88F.PB does not affect the sarcomere structure in pupal indirect flight muscles, with disrupted Z-discs and M-lines.

Expressing LamCUAS.cSa under the control of Scer\GAL4fln.IFM does not affect wing posture and the indirect flight muscles do not seem to show major morphological/ultrastructural defects.

Expressing LamCUAS.cSa under the control of Scer\GAL4HCH.Hand does not affect flight ability and does not seem to cause morphological and physiological defects in the adult heart. 3-week-old cardiomyocytes do not show nuclear envelope, Z-disc or myofibril organization defects.

Expression of LamCScer\UAS.cSa under the control of Scer\GAL4C57 causes no obvious phenotypes and does not affect viability.

The co-expression of LamCUAS.cSa with either AMPKαUAS.cMa, AMPKαK56R.UAS, AMPKαT184D.UAS, AMPKαHMC04979, srlUAS.cRa, srlKK100201, foxoUASp.GFP, foxoKK108485, ThorUAS.cMa, ThorKK108272, ThorLL.UAS, S6kTE.UAS or S6kKQ.UAS, under the control of Scer\GAL4fln.IFM, does not lead to any obvious indirect flight muscle, wing posture or flight defects.

Co-expressing LamCUAS.cSa with either cncdsRNA.UAS.cUa, Keap1KK102175, GstD4GD16151, GstD9KK113031, Atg1UAS.cSa or Atg1K38Q.UAS under the control of Scer\GAL4HCH.Hand causes mild cardiac physiology defects, with cardiomyocytes myofibril organization defects.

Co-expression of LamCScer\UAS.cSa partially rescues the germline stem cell differentiation defect seen in males expressing LamGD1478 under the control of Scer\GAL4tj-NP1624 : 58.9% of testes show strong rescue (although they are shorter than normal), containing germ cells at all stages of differentiation. The remaining 41.1% of testes show weak rescue, showing less severe accumulation of early germ cells but still appearing abnormal.

Comments

Expression of LamCScer\UAS.cSa under the control of Scer\GAL4Act5C.PI rescues head eversion in 13% of LamCLC58 pupae.

Expression of LamCScer\UAS.cSa under the control of Scer\GAL4sr-md710 rescues head eversion in 56% of LamCLC58 pupae. Overall muscle morphology is restored to normal in these pupae.

Expression of LamCScer\UAS.cSa under the control of Scer\GAL4how-24B rescues 89% of LamCLC58 animals to viability.

Expression of LamCScer\UAS.cSa under the control of Scer\GAL4shot-NP3076 rescues 60% of LamCLC58 animals to viability, while a further 23% of the animals show rescued head eversion but die as pupae.
