UAS regulatory sequences drive expression of the C-terminal 141 amino acid residues of BEAF-32 which are tagged at the N-terminal end with a Tag:HA tag and a Tag:NLS(SV40-largeT) nuclear localisation signal. Encodes a dominant negative form of BEAF-32.

Expression of BEAF-32BID.UAS.Tag:HA,Tag:NLS(SV40-largeT) under the control of Scer\GAL4GMR.long causes an increase in the proportion of pale R8 photoreceptors and a reciprocal decrease in the proportion of yellow R8 photoreceptors, compared to controls.

Expression of BEAF-32BID.Scer\UAS.T:Ivir\HA1,T:SV40\nls2 under the control of Scer\GAL4ey.PH results in a rough eye phenotype.

Expression of BEAF-32BID.Scer\UAS.T:Ivir\HA1,T:SV40\nls2 under the control of Scer\GAL4da.G32 can result in lethality, depending on the P{UAS-BEAF-32.BID} insertion line used (the severity of the phenotype depends on the level of expression from the transgene).

Expression of BEAF-32BID.Scer\UAS.T:Ivir\HA1,T:SV40\nls2 under the control of Scer\GAL4ey.PH results in a rough eye phenotype. The severity of the phenotype is increased if the flies carry two copies of BEAF-32BID.Scer\UAS.T:Ivir\HA1,T:SV40\nls2.

Third instar larvae expressing BEAF-32BID.Scer\UAS.T:Ivir\HA1,T:SV40\nls2 under the control of Scer\GAL4AB1 or Scer\GAL434B have smaller salivary glands than normal which are easily fragmented and lack the characteristic well-defined banding pattern.

The mitotic index in the brain is not altered compared to wild type in larvae expressing BEAF-32BID.Scer\UAS.T:Ivir\HA1,T:SV40\nls2 under the control of Scer\GAL4c768. No defects in chromosome condensation are seen, and there is no premature sister-chromatid separation, aneuploidy or anaphase problems. The size of the brain is normal in these larvae.
