Expressing Pink1UAS.cCa under the control of Scer\GAL4Cg.PA does not seem to affect mitochondria size in the third instar larval fat body (as assessed by MitoGFP).

Expressing Pink1UAS.cCa under the control of Scer\GAL4Mef2.PR leads to a small but significant decrease in adult climbing ability; adult thorax muscles do not show obvious signs of degeneration. In 3-day old adults, muscle mitochondria do not show obvious defects; in 35-days old adults, however, mitochondria are slightly smaller, rounder and/or of irregular size, and cristae exhibit mild abnormalities.

The expression of Pink1UAS.cCa under the control of Scer\GAL4Cg.PA does not affect the number of autophagosome-like structures (Atg8a-positive) in the early third instar larval fat body, as compared to controls; in TEM, the mitochondria number and morphology are not dramatically changed, as compared to controls.

The expression of Pink1UAS.cCa under the control of Scer\GAL4GMR.PU does not lead to eye defects (rhabdomere number in both 2- and 30-days old adults), as compared to controls.

The expression of Pink1Scer\UAS.cCa under the control of Scer\GAL4Act.PU in clones within mosaic midguts results in enterocytes exhibiting a significant decrease in the relative mitochondrial content during pupariation, as compared to non-clone enterocytes; expression under the control of Scer\GAL4esg.PU results in a significant decrease in the relative mitochondrial content of midgut intestinal stem cells, from late third instar larval until afer puparium formation, but not at early third instar larval stage or adult stage, as compared to controls.

Expression of Pink1Scer\UAS.cCa under the control of Scer\GAL4GMR.PF at 18[o]C results in a mild rough eye phenotype. A rough eye phenotype is also seen at 29[o]C.

The larval locomotor deficit, very low adult eclosion rate as well as the mitochondrial morphology (enlarged clustered mitochondria) and functional (loss of inner mitochondrial potential measured using cationic fluorescent dye JC-1) defects in adult indirect flight muscles characteristic for animals expressing cluGD13926 under the control of Scer\GAL4Mef2.PR can be suppressed by co-expression of Pink1Scer\UAS.cCa.

Expression of HtrA2Scer\UAS.cYa enhances the rough eye phenotype seen when Pink1Scer\UAS.cCa is expressed under the control of Scer\GAL4GMR.PF at 18[o]C.

Expression of HtrA2S266A.Scer\UAS does not enhance the rough eye phenotype seen when Pink1Scer\UAS.cCa is expressed under the control of Scer\GAL4GMR.PF at 18[o]C.

Expression of HtrA2dsRNA.Scer\UAS.cYa fails to suppress the rough eye phenotype seen when Pink1Scer\UAS.cCa is expressed under the control of Scer\GAL4GMR.PF at 29[o]C.

Comments

Expression of Pink1Scer\UAS.cCa under the control of Scer\GAL4Mef2.PR completely rescues the defects in mitochondrial morphology that are seen in the indirect flight muscles of Pink15 mutants.

Expression of Pink1Scer\UAS.cCa under the control of Scer\GAL4Mef2.PR rescues the mitochondria morphology defects seen in Pink15 mutant indirect flight muscles.
