Only 59% of flies expressing PdfGD299 under the control of Scer\GAL4tim.Scer\UAS remain rhythmic under constant darkness conditions. The remaining rhythmic flies show a significant decrease in the length of the period compared to controls.

Under 12 hour light: 12 hour dark conditions, flies expressing PdfGD299 under the control of Scer\GAL4tim.Scer\UAS show rhythmic locomotor activity, but the evening activity peak is advanced and the morning peak is reduced compared to controls.

Flies expressing PdfGD299 under the control of Scer\GAL4tim.Scer\UAS show normal sleep profiles compared to controls.

Males expressing PdfGD299 under the control of Scer\GAL4Dot.PK (in the presence of Dcr-2Scer\UAS.cDa to enhance RNAi efficiency) show a significant decrease in the amount of (z)-7-Tricosene, (z)-5-Tricosene and (z)-7-Pentacosene compared to wild type during both the subjective day and night (measured on the sixth day of constant darkness).

Expression of PdfGD299 under the control of Scer\GAL4tim.Scer\UAS has no effect on the levels of sex pheromones in males.

Knockdown of Pdf through expression of PdfGD299 under the control of Scer\GAL4tim.Scer\UAS results in a clearly advanced phase of the evening activity peak in LD 12:12 .

Co-expression of ITPGD3825 alters the residual activity rhythms of flies expressing PdfGD299 under the control of Scer\GAL4tim.Scer\UAS under constant darkness conditions: activity is clustered in irregular activity bouts and multiple weak periods ranging from 10 to 35 hours are seen.

Flies co-expressing ITPGD3825 and PdfGD299 under the control of Scer\GAL4tim.Scer\UAS show significantly altered sleep profiles compared to wild type. Total nighttime sleep is decreased, total daytime sleep is decreased, average number of sleep bouts per day is increased, and average sleep bout duration is decreased.

In LD 12:12 , the evening activity peak of npfKK111846/PdfGD299 knockdown flies (under the control of Scer\GAL4tim.Scer\UAS) is different from PdfGD299-knockdown flies: it appears broader and flatter.

Dcr-2Scer\UAS.cDa/Scer\GAL4Mai179/PdfGD299 flies display high levels of arhythmicity under constant darkness. These flies show a normal free-running period under constant darkness and normally phased evening peak under light/dark cycles.
