Scer\GAL4Dot.PK-mediated expression of kirreGD14476 completely abolishes the normal accumulation of ANF-RFP (expressed via Rnor\ANFMhc.T:Disc\RFP) in pericardial nephrocytes.

Adults expressing kirreGD14476 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

The garland cell nephrocytes of third instar larvae expressing kirreGD14476 under the control of Scer\GAL4sns.GCN have shape defects and are more loosely associated than normal.

The numbers of nephrocyte diaphragms in the garland cell nephrocytes of second instar larvae expressing kirreGD14476 under the control of Scer\GAL4sns.GCN is reduced compared to controls, although the NDs that do form appear normal by TEM analysis.

The surface of the garland cell nephrocytes of third instar larvae expressing kirreGD14476 under the control of Scer\GAL4sns.GCN is more uniform than that of control cells when analysed by SEM.

Garland cell nephrocytes from second instar larvae expressing kirreGD14476 under the control of Scer\GAL4sns.GCN show reduced uptake of both dextran 10kD and Alexa Fluor 555 BSA compared to controls after co-incubation with both tracers for 30 seconds.

Garland cell nephrocytes from second instar larvae expressing kirreGD14476 under the control of Scer\GAL4sns.GCN show efficient uptake of dextran 10kD after co-incubation with both Cascade Blue dextran 10kD and fluorescein dextran 500kD for 5 minutes. However, these cells show a very poor uptake of dextran 500kD under these conditions compared to controls.
