    Detailed Description

Neither heterozygous or homozygous Blos1EY06269 has an effect on excitatory junction potential (EJP) amplitude or miniature EJP frequency compared to controls. However both homozygous and heterozygous Blos1EY06269 mutant flies have increased mEJP amplitude and a correspondingly lower quantal content. Blos1EY06269 mutants show a dominant increase in the number of boutons per muscle at the 6/7 synapse, but overall muscle area is unaffected. Homozygous Blos1EY06269 mutant flies show the normal increase in quantal content observed in response to philanotoxin. The rate of vesicle depletion at low frequency stimulation is similar to controls, but rate of vesicle depletion is increased at high frequency stimulation.

Homozygous Blos1EY06269 mutant flies exhibit deficits in short term olfactory habituation.

Blos1EY06269 Dysbe01028 double heterozygotes have no effect on excitatory junction potential (EJP) amplitude or miniature EJP frequency compared to controls. Synaptic vesicles are similar in size to controls. The increase in quantal content normally observed in response to philanotoxin is not seen. One copy of Dysbe01028 suppresses the dominant changes in mEJP amplitude and quantal content seen in Blos1EY06269/+ mutant NMJs. The increase in bouton number seen in Blos1EY06269/+ NMJs is also suppressed.

Df(3L)BSC416 (which deletes Dysb), Blos1EY06269 double heterozygotes have no effect on excitatory junction potential (EJP) amplitude or miniature EJP frequency compared to controls.

Blos1EY06269 Dysbrv double heterozygotes have no effect on excitatory junction potential (EJP) amplitude or miniature EJP frequency compared to controls. One copy of Dysbrv suppresses the dominant changes in mEJP amplitude and quantal content seen in Blos1EY06269/+ mutant NMJs. The increase in bouton number seen in Blos1EY06269/+ NMJs is not suppressed.

Expression of DysbScer\UAS.cDa under the control of Scer\GAL4elav-C155 in a Blos1EY06269/+; Dysbe01028/+ mutant background has no effect on excitatory junction potential (EJP) amplitude or miniature EJP frequency compared to controls. The mEJP amplitude is increased, similar to that seen in Blos1EY06269/+ mutant flies.

One copy of Df(3L)BSC416 suppresses the dominant changes in mEJP amplitude and quantal content seen in Blos1EY06269/+ mutant NMJs.

One copy of Df(3L)BSC416 suppresses the increase in bouton number seen in Blos1EY06269/+ NMJs.

    Comments

Pre-synaptic expression of Blos1Scer\UAS.cCa under the control of Scer\GAL4elav-C155 rescues the changes in mEJP amplitude seen in Blos1EY06269 mutant NMJs.
