Expression of cicKK100838 RNAi under the control of either Scer\GAL4esg-NP5130 or Scer\GAL4Dl.PU (using tub-Gal80[ts] to limit the time of RNAi expression) results in a dramatic overproliferation of intestinal stem cells (ISC) in the adult midgut and increase in ISC mitoses. Expression under the control of Scer\GAL80Su(H).GBE also results in an increase in the number of mitotic cells though majority of these are outside of the Scer\GAL80Su(H).GBE driver expression domain and likely represent a non-autonomous effect of cicKK100838 RNAi on the proliferation of ISCs.

Expression of cicKK100838 under the control of Scer\GAL4esg-NP5130 in intestinal stem cell (ISC) somatic flip-out clones (using tub-Gal80[ts] to limit the time of expression/clone induction) in the adult midgut results in clone overgrowth and highly increased ISC division.

The overproliferation of intestinal stem cells and the increase in the mitotic rate in the adult midgut observed in flies expressing cicKK100838 under the control of either Scer\GAL4esg-NP5130 or Scer\GAL4Dl.PU (using tub-Gal80[ts] to limit the time of RNAi expression) is strongly suppressed by co-expression of pntdsRNA.Scer\UAS.cUa.

The growth of intestinal stem cell somatic MARCM clones expressing cicKK100838 under the control of Scer\GAL4tub.PU in the adult midgut is significantly impaired by combination with pntΔ88 in homozygous state, while combination with either pnt05484Δ33 or pnt07825Δ78 does not reduce the clone growth.
