Expression of jpKK107921 under the control of either Scer\GAL4Act5C.PU or Scer\GAL4twi.PU combined with Scer\GAL4Mef2.PU leads to early lethality, with the Scer\GAL4Mhc.PU driver viable adults do emerge and even have slightly extended lifespan but they also show rapid age-progressive decline in either climbing or flight ability compared to controls, and although their indirect flight muscles do not display any obvious morphological defects in week-old flies, the muscle area is significantly increased in 4-week-old adults and mitochondria therein are smaller and more rounded that in muscles from controls. The diads in the muscles are only rudimentary or have aberrant morphology including a vacuolated sarcomeric reticulum and the adjoining mitochondrial regions also have abnormal cristae morphology.

Scer\GAL4tin.cBa-driven expression of jpKK107921 results in reduced adult lifespan and median survival at 29[o]C compared to controls, and whereas the diastolic interval, systolic interval or heart period length and arrhythmia index in 1-week-old flies at 29[o]C are all comparable to controls, the end-systolic and end-diastolic are significantly increased and the fractional shortening percentage is decreased; the cardiac chambers are enlarged but the thickness of the heart wall is not significantly different from controls. The parallel alignment of transverse myofibrils in the heart tube in the areas surrounding the ostia in jpKK107921-expressing flies is disturbed.

Adult flies expressing jpKK107921 under the Scer\GAL4elav.PU driver have reduced survival rate and display significantly longer recovery times in bang-sensitive analyses at both 1 and 4 weeks after eclosion. Scer\GAL4GMR.PU-driven expression has no obvious morphological phenotype in the adult eye and does not induce depigmentation even in aged flies, but the ommatidia frequently contain fewer photoreceptor cells.

Scer\GAL4rn-GAL4- or Scer\GAL4nub.PU-driven expression of jpKK107921 has no overt effect on adult wing morphology.

The wing vein defects (vein thickening and deltas) characteristic for Dl6B mutants (reared at 29[o]C) are ameliorated by Scer\GAL4nub.PU-driven expression of jpKK107921.

Xenogenetic Interactions
Statement
Reference

The age-progressive retinal degeneration manifested by patchy depigmentation of the adult eye characteristic for flies expressing either Hsap\HTTQ93.ex1.Scer\UAS or Hsap\ATXN3CAG89.Scer\UAS.T:Ivir\HA1,T:Hsap\MYC,T:Zzzz\His6 under the control of Scer\GAL4GMR.PU is exacerbated by co-expression of jpKK107921.

Complementation and Rescue Data
Comments

The adult eye phenotype (disrupted ommatidial lattice, supernumerary interommatidial bristles) induced by Scer\GAL4GMR.PU-driven expression of jpUAS as well as the reduced wing size observed upon expression with Scer\GAL4nub.PU is restored by co-expression of jpKK107921.
