Description

One of the two UAS regulatory elements present in the P{XP} element of the P{XP}jpd04563 insertion has been removed by excising the FRT cassette. The remaining UAS regulatory element is in the correct orientation to drive overexpression of jp in the presence of a GAL4 driver.

Expression of jpUAS under the control of Scer\GAL4Mhc.PU yields viable adult flies with slightly reduced lifespan that also show rapid age-progressive decline in either climbing or flight ability compared to controls, although their indirect flight muscles (and mitochondria therein) do not display any obvious morphological defects even in aged flies. However, the diads in the muscles have aberrant diads with elongated sarcomeric reticulum and occasionally show abnormal positioning away from the myofibrils and embedded in the mitochondria.

Scer\GAL4tin.cBa-driven expression of jpUAS results in reduced adult lifespan and median survival at 29[o]C compared to controls, and whereas the diastolic interval, systolic interval or heart period length and arrhythmia index in 1-week-old flies at 29[o]C are all comparable to controls, the end-systolic and end-diastolic are significantly increased and the fractional shortening percentage is decreased; the cardiac chambers are enlarged but the thickness of the heart wall is not significantly different from controls. The parallel alignment of transverse myofibrils in the heart tube in the areas surrounding the ostia in jpUAS-expressing flies is disturbed.

Adult flies expressing jpUAS under the Scer\GAL4elav.PU driver have reduced survival rate and display significantly longer recovery times in bang-sensitive analyses at 4 weeks after eclosion (but not at 1 week post-eclosion). Scer\GAL4GMR.PU-driven expression leads to mildly disrupted ommatidial organization with some misshapen ommatidia and supernumerary bristles, the ommatidia also frequently contain excessive photoreceptor cells, but no depigmentation is observed even in aged flies.

Scer\GAL4Act5C.PU-driven expression of jpUAS results in supernumerary vibrissae and sternopleural bristles, duplication of the anterior scutellar macrochaetae and wing vein deltas at the wing margin. Expression driven by Scer\GAL4rn-GAL4 or Scer\GAL4nub.PU also induces formation of wing vein deltas along with thickening of longitudinal veins and overall decrease in the wing blade size.
