The testes of newly eclosed flies that have expressed chicKK112358 throughout development under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 completely lack germline stem cells. In contrast to wild type testes, elongated spermatid bundles are located close to the apical tip. Spermatogonia and spermatocytes are rare or not detected.

When chicKK112358 is expressed in somatic cells under the control of Scer\GAL4C587 from eclosion onwards the testes germ cell overproliferate at the expense of proliferation: at day one after expression induction all stages of spermatogenesis are observed, but by days 4-5 spermatocytes and mature spermatids are markedly reduced in number and the testes are filled with clusters of small cells. These small cells have branched fusomes and express the bam differentiation factor, indicating that they are transit-amplifying spermatogonia rather than GSCs or gonialblast-like cells. In contrast to wild type, where mitotic cells are only seen close to the apical tip in GSCs, 2-, 4- and 8-cell cysts, in testes expressing chicKK112358 mitotic cells are seen in larger clusters far from the apical tip. Expression of chicKK112358 under the control of Scer\GAL4C587 also affects the somatic cell lineage: by 4-5 days after expression induction somatic cyst stem cells are mostly absent. Cyst cells are decreased in number and sometimes exhibit abnormal morphology: nuclei appear larger than normal and have de-condensed chromatin. The cytoplasmic extensions that normally surround the germline cells are not observed.

Expression of chicKK112358 under the control of Scer\GAL4e22c results in a failure of wound closure, with open wounds being present in larvae 24 hours after wounding.
