Description

Imprecise excision of the progenitor insertion has resulted in an 800bp deletion, removing the promoter and extending into the first intron of Ada2b.

Ada2bd842 follicle cell clones frequently show picnotic nuclei or frequently fail to show the expected gene amplification (BrdU-positive foci) normally observed in controls.

Homozygotes undergo apparently normal development until the P5 stage, except that the larval and first stages of pupal development take slightly longer to complete than in their control siblings. At the P5 stage, the legs and head show reduced development compared to controls and 85% of the animals die. A small fraction of the mutants reach the pharate adult stage.

Homozygotes and Ada2bd842/Df(3R)Ada2bd52 transheterozygotes show 77% lethality at the P5 pupal stage, and only 23% reach the pharate adult stage. No animals emerge as adults.

In disc-derived body-parts, homozygous clones develop as frequently as control clones, but are significantly smaller than control clones. Homozygous clones on the abdomen are as large as control clones.

Heterozygous adults show a reduction in eye pigmentation compared to wild type.

Mutant imaginal discs exhibit significantly lower levels of apoptosis (as stained by Acridine Orange) when subjected to X-ray irradiation (with 4,000R) compared to wild-type discs.

The puff in the 61C region induced by Scer\GAL4hs.PB-mediated expression of P{UAS-Fcp1.dsRNA.WIZ}61C remains readily detectable in Ada2bd842 homozygotes.

The presence of p53Scer\UAS.Ex, under the control of Scer\GAL4GMR.PF does not affect the reduction in eye pigmentation observed in Ada2bd842 heterozygotes.

10% of Ada2bd842 mutants carrying a single copy of Ada2bScer\UAS.P\T.S.T:Avic\GFP-EGFP emerge as adults.

85% of Ada2bd842 mutants carrying a single copy of Ada2bScer\UAS.P\T.T:Avic\GFP-EGFP emerge as adults.

40% of Ada2bd842 mutants expressing Ada2bScer\UAS.S under the control of Scer\GAL4da.G32 emerge as adults.

Expression of Ada2bScer\UAS.cVa under the control of Scer\GAL4Act.PU partially rescues the pupal P5 stage lethality of Ada2bd842 animals, supporting development to the pharate adult (P14) stage in 80% of cases.

Expression of Ada2a::Ada2b2a2b.Scer\UAS under the control of Scer\GAL4Act.PU partially rescues the pupal P5 stage lethality of Ada2bd842 animals, with over 50% of the animals reaching the pharate adult (P14) stage. A small fraction of animals emerge as adults, but they die within 12 hours of hatching, with their wings still folded.

Expression of either Ada2a::Ada2b2bS.2a.Scer\UAS, Ada2a::Ada2b2bM.2a.Scer\UAS, Ada2a::Ada2b2bL.2a.Scer\UAS or Ada2aScer\UAS.cVa under the control of Scer\GAL4Act.PU fails to rescue the pupal P5 stage lethality of Ada2bd842 animals.

Ada2bScer\UAS.P\T.T:Avic\GFP-EGFP is able to produce an 80% phenotypic rescue of Ada2bd842 mutants in the absence of a Scer\GAL4 driver. When expression of Ada2bScer\UAS.P\T.T:Avic\GFP-EGFP is driven by Scer\GAL4Act5C.PU the same degree of rescue of Ada2bd842 is seen as in the absence of the driver.
