The expression of mstJF03280 under the control of Scer\GAL4tub.PU or Scer\GAL4da.PU induces lethality during development.

The expression of mstJF03280 under the control of Scer\GAL4Mef2.PU leads to short-lived adults, who exhibit a significant decrease in food intake, a near absence of gut contractions, a decreased movement of food through the intestinal tract (and a near absence of fecal deposition), a progressive abdominal dilation, a progressive and significant intestine dilation, a severe increase in gut permeability, increased signs of infection (i.e. melanization marks), and progressively and severely disturbed circular and longitudinal intestinal muscle fibers, associated with a disrupted sarcomere structure and weakened actin filaments, but no changes in sensitivity to food (measured by the frequency of proboscis extension responses to sucrose), as compared to controls; the visceral muscle fibers of old individuals also exhibit a significant increase in apoptosis and disrupted mitochondria structure, as compared to controls. Expression under the combined control of Scer\GAL4Mef2.PU and Gal80[ts] (for adulthood-only expression) or Scer\GAL4Mef2.PU and Scer\GAL80nSyb.PS still leads to a dilated intestine, as compared to controls.

Adults expressing mstJF03280 under the control of Scer\GAL4how-24B leads to short-lived adults, who exhibit a significant decrease in food intake, a dilated intestine, a near absence of gut contractions, and severely disturbed circular and longitudinal intestinal muscle fibers, as compared to controls. Expression under the control of Scer\GAL4tey-5053A leads to a small but significant increase in food intake, a mildly thinner intestine and to a moderately disturbed longitudinal (but not circular) intestinal muscle fibers, as compared to controls. Expression under the control of Scer\GAL4Myo31DF-NP0001 has no significant effect in feeding behavior, in intestine diameter or in circular and longitudinal intestinal muscle fibers, as compared to controls.

The expression of mstJF03280 under the control of Scer\GAL4Act5C.PI, but not Scer\GAL4Cg.PA or Scer\GAL4nSyb.PS, leads to adults exhibiting a significant decrease in food intake, as compared to controls.

The expression of mstJF03280 under the control of Scer\GAL4Mhc.PU has no obvious effect on the locomotor activity, intestine diameter or skeletal muscle structures in adults, as compared to controls.

The lethality induced by the expression of Act42AScer\UAS.P\T.T:Avic\GFP, Act57BScer\UAS.P\T.T:Avic\GFP, Act87EScer\UAS.P\T.T:Avic\GFP, Act88FScer\UAS.P\T.T:Avic\GFP, or Act5CScer\UAS.P\T.R.T:Avic\GFP under the control of Scer\GAL4Mef2.PU is not suppressed by the co-expression of mstJF03280.

The co-expression of Act79BScer\UAS.P\T.T:Avic\GFP fully suppresses the dilated intestine and the visceral muscle actin defects and partially suppresses the decreased lifespan observed in adults expressing mstJF03280 under the control of Scer\GAL4Mef2.PU.

Xenogenetic Interactions
Statement
Reference

The decreased lifespan, the decreased food intake, the decreased gut contractions, the dilated intestine, and the visceral muscle structure defects (including in actin filaments) observed in adults expressing mstJF03280 under the control of Scer\GAL4Mef2.PU are rescued by the co-expression of BacA\p35Scer\UAS.cHa.

Comments

The decreased lifespan, the decreased food intake, the decreased gut contractions, the dilated intestine, and the visceral muscle structure defects observed in adults expressing mstJF03280 under the control of Scer\GAL4Mef2.PU are rescued by the co-expression of mstScer\UAS.ORF.

The decreased lifespan observed in adults expression of mstJF03280 under the control of Scer\GAL4Mef2.PU is fully rescued by the co-expression of mstΔ2.Scer\UAS.T:Ivir\HA1, is partially rescued by the co-expression of mstScer\UAS.T:Avic\GFP, and is not rescued by the co-expression of either mstΔ1.Scer\UAS.T:Ivir\HA1 or mstΔ3.Scer\UAS.T:Ivir\HA1.
