Detailed Description
Statement

Expressing Pink1JF01672 under the control of Scer\GAL4Cg.PA leads to mitochondrial defects in the third instar larval fat body, including enlarged, fragmented and/or clustered mitochondria (as assessed by MitoGFP).

Expressing Pink1JF01672 under the control of Scer\GAL4Mef2.PR leads to a significant decrease in adult climbing ability.

The expression of Pink1JF01672 under the control of Scer\GAL4Act.PU in clones within mosaic midguts results in enterocytes during pupariation exhibiting a significant increase in the transverse cell area, a significant decrease in the relative amount of autophagosomes, as assessed by a Atg8a fluorescent reporter, and a significant increase in the relative mitochondrial content, as compared to non-clone enterocytes; however, no significant change in the relative mitochondrial content is observed at 4 hours after puparium formation, as compared to non-clone enterocytes.

The increased relative mitochondrial content and the decreased relative amount of autophagosomes observed during pupariation in enterocytes within midgut clones expressing Pink1JF01672 under the control of Scer\GAL4Act.PU are suppressed by the clonal co-expression of Hsap\PARK2Scer\UAS.cYa, even leading to a significant decrease in relative mitochondrial content and a significant increase in relative amount of autophagosomes, as compared to non-clone enterocytes.
