Expression via "HHLT-gal4" results in loss of progenitor cells in the primary lobes of the larval lymph gland. ("HHLT-gal4" is a combination of Scer\GAL4HCH.Hand and Scer\GAL4Hml.Δ that express in early lymph gland blood cells (hemocytes) and circulating and sessile blood cells, respectively, and incorporates Scer\GAL4Scer\FRT.Rnor\CD2.Act5C to maintain expression in the lymph gland after Scer\GAL4HCH.Hand is down-regulated.)

Adult males and females expressing GaleNIG.12030R under the control of Scer\GAL4Act5C.PI from the late larval or early-to-mid-pupal stage onwards (the temperature sensitive Scer\GAL80ts.αTub84B allele is used to regulate the timing of expression) show a dramatic reduction in lifespan compared to control flies in the presence of 175mM galactose.

100% of Scer\GAL4drm.7.1, GaleNIG.12030R die at 28-29{o]C.

GaleNIG.12030R, Scer\GAL4Act5C.PI, Scer\GAL80ts.αTub84B flies show progressively milder phenotypes as animals are shifted to the restrictive temperature later in development: animals shifted at embryonic, larval or early pupal stages die; animals shifted later in pupation produce viable adults with negative effects on life span, fertility and female fecundity.

Expression of Zzzz\wbgUScer\UAS.cDa under the control of Scer\GAL4Act5C.PI rescues the lethality caused by expression of GaleNIG.12030R under the control of Scer\GAL4Act5C.PI, but the rescued males show loss of fecundity (produce no offspring when mated to wild-type females). The rescued females show loss of fecundity (produce no offspring when mated to wild-type males) if expression occurs at third instar larval stages or earlier but show normal fecundity if expression is at the pupal stage at days 18-19.

Expression of Ecol\galEScer\UAS.cDa under the control of Scer\GAL4Act5C.PI rescues the lethality caused by expression of GaleNIG.12030R under the control of Scer\GAL4Act5C.PI. The rescued males show normal fecundity (number of offspring produced when mated to wild-type females). The rescued females show reduced fecundity (reduced number of offspring produced when mated to wild-type males) if expression occurs at the pupal stage at days 11-17 or earlier but show normal fecundity if expression is at the pupal stage at days 18-19.

Expression of Hsap\GALEScer\UAS.cSa under the control of Scer\GAL4Act5C.PI rescues the lethality caused by expression of GaleNIG.12030R under the control of Scer\GAL4Act5C.PI. The rescued males and females show normal fecundity (number of offspring produced when mated to wild-type flies of the opposite sex).

The short lived phenotype seen in adult males and females expressing GaleNIG.12030R under the control of Scer\GAL4Act5C.PI from the late larval or early-to-mid-pupal stage onwards (the temperature sensitive Scer\GAL80ts.αTub84B allele is used to regulate the timing of expression) in the presence of 175mM galactose is completely suppressed by expression of Hsap\GALEScer\UAS.cSa under the control of Scer\GAL4Act5C.PI.

Expression of Hsap\GALEScer\UAS.cSa using Scer\GAL4Act5C.PI rescues the lethality of GaleNIG.12030R, Scer\GAL4Act5C.PI animals. Rescued animals are viable, fertile, and appear morphologically normal.
