The distribution and dendritic projections of Ir84a-expressing neurons, as revealed by Mmus\Cd8aScer\UAS.T:Avic\GFP, is unaffected in Ir84aGAL4 homozygotes.
The >50 spikes per second seen in ac4 sensilla in response to phenylacetic acid and phenylacetaldehyde is completed abolished in Ir84aGAL4 homozygotes. The basal activity in Ir84aGAL4 ac4 sensilla is also lower than in controls.
In single-pair courtship assays, Ir84aGAL4 males court wild type females significantly less than do wild type males; most individual components of the courtship ritual are affected. Homosexual courtship is also markedly reduced in Ir84aGAL4 males. By contrast, Ir84aGAL4 females do not show overt defects in reproductive behaviours.
The increased courtship seen in wild type males when killed female objects are perfumed with phenylacetic acid (or fly food) is not observed in Ir84aGAL4 males.
Ir84aGAL4 is rescued by Scer\GAL4Ir84a-GAL4/Ir84aUAS.cBa
Ir84aGAL4 is partially rescued by Scer\GAL4Ir84a-GAL4/Ir84aUAS.cBa
Expression of Ir84aScer\UAS.cBa or Ir84aScer\UAS.T:Hsap\CALR,T:Avic\GFP-EGFP using Scer\GAL4Ir84a-GAL4 restores the response to phenylacetic acid and phenylacetaldehyde in Ir84aGAL4 homozygotes. Ir84aScer\UAS.cBa expression also rescues the reduced courtship seen in Ir84aGAL4 males.
The increased courtship seen in wild type males when killed female objects are perfumed with phenylacetic acid (or fly food) is not observed in Ir84aGAL4 males, but this is partially restored by expression of Ir84aScer\UAS.cBa.
