Description

UAS drives expression of the 2n4r isoform of Hsap\MAPT. (This construct has more 5' and 3' UTR sequence than P{UAS-MAPT.VTR}.)

Expression of Hsap\MAPTScer\UAS.B under the control of Scer\GAL4GMR.PU results in a disruption of the eye ommatidial lattice in adults.

Pan-neuronal expression of Hsap\MAPTScer\UAS.B under the control of Scer\GAL4elav.PU results in severe mushroom body defects.

Temperature shift experiments reveal that conditional expression of Hsap\MAPTScer\UAS.B under the control of Scer\GAL4elav.PU and Scer\GAL80ts.αTub84B during the first 4 h of embryogenesis at 19[o]C does not affect adult MB structure. In contrast, continued expression of Hsap\MAPTScer\UAS.B 8 h into embryogenesis results in MB abnormalities in adults. Continued expression of Hsap\MAPTScer\UAS.B up to 14 h after egg laying does not result in more severe deficits.

Expression of Hsap\MAPTScer\UAS.B under the control of Scer\GAL4elav.PU results in severe and specific mushroom body defects.

Associative olfactory learning is nearly abolished in adult animals expressing Hsap\MAPTScer\UAS.B under the control of Scer\GAL4elav.PU.

The defective eye morphology (disrupted ommatidial lattice) observed in adult flies expressing Hsap\MAPTScer\UAS.B under the control of Scer\GAL4GMR.PU is significantly worsened by co-expression of Rho1dsRNA.Scer\UAS RNAi with necrotic spots appearing in the posterior eye. Combination with CG11739d06383 also enhances the aberrant eye phenotype as the eye size is much reduced, whereas co-expression of PgmHMS01333 RNAi has no apparent effect.
