Scer\GAL4nos.UTR.T:Hsim\VP16-mediated expression of eIF4G2GL00469 does not appear to affect spermatocyte cyst development, but the elongated spermatids in the late stages of differentiation that encompass the length of the wild-type testes are absent. Meiotic and post-meiotic stage cysts are observed, but the spermatid cysts appear to be arrested in the early elongation phase and degenerate thereafter. Multiple nuclei in haploid onion-stage spermatids are observed, which appear larger than the wild-type cysts. In contrast, nuclear condensation during the mitotic and meiotic phases is not affected, and mitochondrial differentiation appears normal.
Testes expressing eIF4G2GL00469 via Scer\GAL4bam.T:Hsim\VP16 have no detectable meiotic or post-meiotic elongated spermatid cysts. Instead, the testes are mostly filled with large mature spermatocytes which degenerate at the distal tip. Later spermatid stages with elongated flagella are absent - mature primary spermatocytes are arrested at the G2/M transition. In contrast to wild-type, the nuclei contain dispersed chromatin and the mitochondria failed to fuse fully. Cytokinesis is normal.

Males expressing both eIF4GHMS00762 and eIF4G2GL00469 via Scer\GAL4nos.UTR.T:Hsim\VP16 are sterile and produce rudimentary testes. The apical tips are filled with masses of dividing cells, indicating an arrest before the primary spermatocyte stage.
