Expression of tumHMS01417 RNAi under the control of Scer\GAL4Act5C.PU (using tub-Gal80[ts] to limit the RNAi expression to the adult stage) results in germline stem cell (GMC) loss and premature differentiation into spermatogonial cells in adult testes. Expression under Scer\GAL4nos.UTR.T:Hsim\VP16 also results in GMC loss, whereas expression under Scer\GAL4C587 leads to GMC differentiation defects. Scer\GAL4bam.T:Hsim\VP16-driven expression gives rise to polyploid cells in the testes.
