Adults expressing parkHMS01651 under the control of Scer\GAL4Act5C.PI or Scer\GAL4ple.PF show a progressive decline in climbing activity, and a progressive decrease in DA neuron number (PPL1, PPL2, PPM1/2 and PPM3 clusters); DA neurons also show decreased abundance of mitochondria.

The expression of parkHMS01651 under the control of Scer\GAL4Act.PU in clones within mosaic midguts results in enterocytes during pupariation, but not after puparium formation, exhibiting a significant increase in the transverse cell area, a significant increase in the relative mitochondrial content and a significant decrease in the relative amount of autophagosomes, as assessed by a Atg8a fluorescent reporter, as compared to non-clone enterocytes.

Expression of parkHMS01651 under the control of Scer\GAL4tin.CΔ4 evokes heart tube hypocontractility at one and four weeks after eclosion, and increases cardiomyocyte mitochondrial size. The lifespan of these flies is similar to controls but the flies display an accelerated age-related decline in climbing performance. Heart tube end-systolic and end-diastolic dimensions are increased and mitochondria have decreased DNA, increase mitochondrial reactive oxygen species (ROS) and increased numbers of depolarized organelles.

However, the mitochondria show characteristics of both parent lines: they are still enlarged as is seen in parkHMS01651, and appear fragmented as is seen with MarfmiRNA.cUa.Scer\UAS.
