Homozygous Gr5aLEXA mutant flies exhibit a reduced proboscis extension response to maltose and trehalose upon stimulation of either the labial or tarsal taste sensilla, and several other sugars upon stimulation of the tarsal sensilla.

Gr61a1 Gr5aLEXA Df(3L)ΔGr64 mutants expressing P{t15.3.R1} and P{UAS-Rescue.R2} (i.e. lacking all 8 sweet gustatory receptor genes) do not exhibit any defects in fructose preference in two-choice preference assays.

Adult females homozygous for Gr5aLEXA, Gr61a1 and Df(3L)ΔGr64, or with Gr5aLEXA/+, Gr61a1/+ and Gr64fLEXA/Df(3L)ΔGr64, have a significant increase in preference for softer 100 mM sucrose-containing, but not plain, agarose for egg laying, compared to controls; these mutants also have a significant reduction in preference for 100 mM sucrose-containing (rather than plain) agarose for egg laying, compared to controls.

Adults with homozygous Gr5aLEXA, Gr61a1 and Df(3L)ΔGr64 have proboscis extension rates that are significantly reduced in response to sucrose and significantly increased in response to some fatty acids, compared to controls.

Gr5aLEXA, Gr61a1, Df(3L)ΔGr64 triple homozygotes that also bear P{t15.3.R1} (P{t15.3.R1} rescues the lethality of Df(3L)ΔGr64 homozygosity) have impaired responsiveness to sweet chemical: sweet taste 5b sensilla GRNs virtually lack the expected responsiveness to the chemicals Sucrose, Fructose, Maltose, Trehalose, Glucose, Melezitose, Arabinose and Glycerol, as compared to controls; sweet taste 5v sensilla GRNs show the expected responsive only to Sucrose and Fructose, but lack responses to Maltose, Trehalose and Glucose.

Expressing Gr64aUAS.cUa under the control of Scer\GAL4Gr61a.4.9 restores the response of 5v sensilla GRNs only to Maltose but does not affect the responses of 5b sensilla GRNs.

Expressing Gr61aUAS.cUa under the control of Scer\GAL4Gr61a.4.9 restores a moderate response of 5v sensilla GRNs only to Glucose but does not affect the responses of 5b sensilla GRNs.

Expressing Gr5aUAS.cUa under the control of Scer\GAL4Gr61a.4.9 restores a small response of 5v sensilla GRNs only to Trehalose but does not affect the responses of 5b sensilla GRNs.

Co-expressing Gr64aUAS.cUa and Gr64eUAS.cUa under the control of Scer\GAL4Gr61a.4.9 restores the response of 5b sensilla GRNs only to Maltose and Sucrose, but does not affect the responses by 5v sensilla GRNs.

Co-expressing Gr64bUAS.cUa and Gr64eUAS.cUa under the control of Scer\GAL4Gr61a.4.9 restores the response of 5b sensilla GRNs only to Glycerol, but does not affect the responses by 5v sensilla GRNs.
