Description

Duplication at the p53 locus. The 5' copy consists of wild-type exons 1 to 6, a partial insertion of pTV2 vector sequence, another copy of exon 6 and part of exon 7. The 3' copy lacks exon 1 and part of exon 2. A w+mW.hs marker and a single FRT site are located between the two copies.

γ-irradiated p53-ns/p53TM3 embryos and larval wing discs (the Tm3, Sb balancer has an inversion break point at the p53 gene), as well as γ-irradiated p53-ns homozygous wing discs, fail to show the expected increase in apoptosis.

Homozygous Dp(3;3)p53-ns mutant flies (comprised of the p53-ns.5' and p53-ns.3' alleles) mutant flies exhibit a reduction in survival following acute starvation during early adulthood (1-2 days). Reduced survival rates are seen in both males and females and also in 5-7 day old flies subjected to starvation. Similar effect on survival rates are seen in young adult flies or larvae exposed to complete nutrient deprivation, as well as larvae exposed to diluted food. Triacylglycerides (TAGs) and glycogen are used at higher rates in p53-ns.3' mutant flies exposed to nutrient deprivation compared to controls.

One copy of p53-ns suppresses the increase in survival rate in response to starvation seen in adult Dcr-1Q1147X/+ mutant flies.
