Ectopic expression of SLC22AScer\UAS.cGa in all mushroom body neurons using either the Scer\GAL4238Y or the Scer\GAL4GMR13F02 driver results in significant 3hr memory loss in an olfactory classical conditioning assay in adult flies.

Odor (Benzaldehyde and Octanol) and shock (electrical) avoidance was normal (tested with Scer\GAL4238Y and Scer\GAL4GMR13F02).

A conditional ectopic expression of SLC22AScer\UAS.cGa after eclosion in all mushroom body neurons using the Scer\GAL4238Y driver (with GAL80ts to limit the stage of expression) results in loss of 3hr memory in an olfactory classical conditioning assay whereas expression during development (before eclosion) has no effect on memory test performance in adults.

Supplementing food with dopamine or histidine impairs 3hr memory of both control adult flies and of the already memory-defective flies expressing SLC22AScer\UAS.cGa under the control of the Scer\GAL4238Y driver in an olfactory classical conditioning assay.

Supplementing food with choline impairs 3hr memory of control adult flies but improves that of the already memory-defective flies expressing SLC22AScer\UAS.cGa under the control of the Scer\GAL4238Y driver in an olfactory classical conditioning assay.

Co-expression of SLC22AScer\UAS.cGa with SLC22AKK112539 RNAi using either Scer\GAL4238Y or Scer\GAL4GMR13F02 driver fully rescues the 3hr enhanced memory phenotype in adult flies in an olfactory classical conditioning assay produced by the SLC22 knock-down.
