PRL-12/Df(2L)r10 transheterozygotes show delayed hatching. Adults have apparently normal size and morphology, but occasionally show held-up wings; they display climbing defects and are unable fly. Their posterior dorsocentral bristle neurons show normal neuron cell body size and morphology, and normal axon caliber, but the contralateral branch covers significantly less territory, as compared to controls; the ventral nerve cord is significantly smaller than in controls, particularly in the metathoracic neuropil; except for slightly shorter posterior projections, due to a significantly smaller VNC, the projections of SC mechanosensory neurons are normal.

In PRL-12 homozygous adults, the posterior dorsocentral bristle neurons show loss of synaptic arborizations in the contralateral axon collateral, and the antennal lobes and mushroom bodies are disorganized, with evident axonal or synaptic defects (e.g. ORN target area appears disrupted/less homogenous and the synaptic termination area is smaller despite of a comparable number of Or83c-positive neurons; mushroom body fusion, with midline crossing of some or all mushroom body axons), as compared to heterozygous controls. The third instar larval neuromuscular junction appears normal. At 72h after puparium formation there are significantly fewer satellite growth cones per axon segment, which extend fewer filopodial protrusions, but no reduction of filopodia emanating from the main branch, as compared to controls; these defects are not observed at 47h and 90h after puparium formation.

PRL-12/PRL-16 posterior dorsocentral bristle neurons show a significant decrease in the number of synapses (Brp-positive puncta) in the contralateral branch, but not in the anterior branch, as compared to controls; mechanosensory neurons show a significant decrease in terminal arbor territory, as compared to controls.

In PRL-12/PRL-14 adults, the ventral nerve cord show comparable cell numbers to controls, and the posterior dorsocentral bristle neurons do not show striking differences in synapse ultrastructure, as compared to controls.

Posterior dorsocentral bristle neurons of individuals that are both PRL-12/PRL-14 transheterozygous and express PRL-1UAS.cPa under the control of Scer\GAL4pnr-MD237 do not show significant projection defects (terminal arbor territory or number of synapses/ Syt1-positive puncta) in the contralateral branch, as compared to controls.
